Summary: This systematic review with meta-analysis evaluates the utility of pharmacogenetic tests (PGTs) in the routine clinical treatment of mood disorders. The results indicate that PGT suggestions can lead to better efficacy outcomes for patients with major depressive disorder (MDD), but not for patients with bipolar disorder (BD). However, PGTs show promising results in terms of setting up more tolerated therapies. Overall, PGTs are important in supporting clinicians' choices in psychopharmacological therapies.

Summary: The antidepressant effects of memantine in bipolar depression are promising, with both experimental and clinical evidence supporting its efficacy. The presented case study demonstrates that memantine can be an effective and safe adjunct treatment for mood stabilization in bipolar depression. This suggests that memantine may offer a new approach for patients who do not respond well to traditional medications.

Summary: In this study, adjunctive antidepressant treatment was not associated with lower depressive symptoms or higher mania symptoms in outpatients with bipolar disorder. There were no differences in treatment effects on mania scales, and potential biases due to nonrandomized design complicated causal interpretations, with no evidence suggesting better treatment effects of adjunctive antidepressants.

Summary: This study compared the socio-demographic and clinical features of bipolar disorder patients treated vs. not treated with antidepressant drugs (ADs). The results showed that patients treated with ADs were older, more likely to be retired, and more likely to have had a first major depressive episode. Patients receiving ADs also had more affective episodes and longer illness duration compared to those not treated with ADs.

Summary: Studies have shown that there is a positive relationship between resistance to antidepressants and the progression of bipolar disorder. However, the specific influence of different types of antidepressants, such as SSRIs and SNRIs, in this context has not been thoroughly investigated. This study recruited a total of 5,285 young people with antidepressant-resistant depression and 21,140 with antidepressant-responsive depression. It was found that those with resistance to antidepressants, especially to both SSRIs and SNRIs, were at an increased risk of developing bipolar disorder.

Summary: This study aimed to investigate the genomics underlying the enlarged volume of the hippocampus in bipolar disorder patients after long-term lithium administration. The results suggest that lithium may promote the generation of new neurons and their differentiation into neuroblasts, neurons, or microglia, leading to hippocampal enlargement. Several genes, including CCND1, LOXL2, and PRNP, were identified as being involved in the drug response and chronic effects of lithium in hippocampal cells. GSK-3β was also found to be a key hub in the pharmacogenomic network of lithium.

Summary: The study investigated the possible association of polygenic risk scores (PRSs) for bipolar disorder, major depressive disorder (MDD), neuroticism, and schizophrenia (SCZ) with antidepressant non-response or non-remission in patients with MDD. Methodological improvements in PRS calculation may enhance their predictive performance in precision psychiatry.

Summary: This study aimed to explore the association between thyroid autoimmunity and treatment-emergent mania (ATEM) in pediatric mood disorders. The findings showed that the seroprevalence and titer of anti-thyroid peroxidase antibodies (TPO-abs) were significantly higher in patients with ATEM compared to controls. Logistic regression analysis revealed a significant association between TPO-abs seroprevalence and ATEM even after adjusting for various factors. These findings suggest that TPO-abs could potentially serve as a biomarker for assessing the risk of ATEM in children and adolescents.

Summary: This study retrospectively evaluated the applicability and progression of four staging models in a sample of 100 bipolar disorder patients. The results showed a pattern of stage worsening for each model, with a significant increase at each time point. Patients with lower education level, younger age at first episode, shorter illness duration, and shorter duration of untreated illness showed greater stage increases. Lower stage values were associated with certain characteristics, such as BD II, no psychiatric hospitalization, depressive onset, and few lifetime episodes.

Summary: This study compared the efficacy and discontinuation of augmentation agents in adult patients with treatment-resistant depression through a systematic review and network meta-analyses. The findings suggest that certain adjunctive atypical antipsychotics, thyroid hormones, dopamine compounds, and lithium are superior in terms of efficacy. However, ziprasidone, mirtazapine, and cariprazine had higher discontinuation rates. Further research and head-to-head studies are needed to strengthen treatment options for treatment-resistant depression.

TD+ participants were older, more likely to be female, and have type I bipolar illness compared to the TD- group. Limitations of the study included a cross-sectional design and the lack of severity assessment for tardive illness.

Summary: Bipolar disorder with co-occurring attention deficit-hyperactivity disorder (ADHD) is associated with a younger age of onset, male predominance, higher rates of comorbid anxiety and substance use disorders, and poorer response to mood stabilizing treatments. Genetic analysis shows that all subsets of bipolar disorder patients have higher genetic risk for both bipolar disorder and ADHD, with BD+cADHD patients having a higher ADHD genetic risk.

Summary: This study investigated the relationship between mitochondrial function and the risk of treatment-emergent mania in patients with bipolar disorder. Summary: Insulin resistance (IR) is associated with poorer clinical outcomes and inadequate treatment response in patients with bipolar disorder (BD). BD patients with IR exhibit impairments in verbal memory and executive function, as well as smaller hippocampal volumes and prefrontal neurochemical alterations. They are also more likely to have chronic and rapid cycling courses and lower Global Assessment of Functioning scores. BD patients with impaired glucose metabolism also have a higher rate of poor response to mood stabilizers.

Summary: This study examined the relationship among family accommodation (FA), posttraumatic stress symptoms (PTSS), and functional impairment. The results showed that FA was significantly associated with PTSS and functional impairment. Baseline FA partially mediated the relationship between baseline PTSS and functional impairment. Changes in FA from pre- to post-treatment were associated with relevant outcome variables at post-treatment and 12-month follow-up.
