Summary: Itraconazole significantly reduces the development of esophageal adenocarcinoma and SHH expression in a preclinical animal model of Barrett's Metaplasia. In humans, BE tissue expresses higher levels of SHH, Indian Hedgehog, and bone morphogenic protein compared to normal squamous esophageal epithelium.

Summary: This study aims to establish a prognostic model based on extragastric lymph node metastasis (ELNM) and lymph node ratio (LNR) in node-positive gastric cancer (GC). The results show that this model has good prognostic prediction in patients with node-positive GC.

Summary: The expression of ALDOA in gastric cancer is associated with the progression and prognosis of the disease. High ALDOA expression is correlated with poor prognosis and lymph node metastasis. ALDOA may impact the progression of gastric cancer by affecting the expression of immune molecules. Therefore, ALDOA could serve as a promising biomarker and therapeutic target for evaluating the prognosis of gastric cancer.

Summary: COPD is a chronic airway disease that affects millions of people worldwide and is ranked as the third leading cause of death. Genetic susceptibility, environmental factors, and the combined effects of these factors play important roles in the development of COPD. The Hedgehog signaling pathway and its inhibitor HHIP are potential targets for therapeutic interventions in COPD.

Summary: The study reveals the previously unrecognized role of the endogenous hedgehog signaling pathway in regulating pathogenic inflammation within the central nervous system. By targeting CD4 T cells and modulating the production of inflammatory cytokines, the hedgehog pathway shows potential as a therapeutic strategy to limit the progression of ongoing neuroinflammation. Systemic administration of a hedgehog agonist after disease onset effectively halts disease progression and reduces neuroinflammation and associated neuropathology.

Summary: This study reveals that the activation of the Shh-Gli-Bmi1 signaling pathway plays a critical role in the bone anabolic action induced by p27 deficiency. Bmi1 may be an important therapeutic target for treating osteoporosis caused by the activation of p27 signaling or inactivation of sonic hedgehog signaling.

Summary: The geriatric nutritional risk index (GNRI) is an important determinant of overall survival (OS) and disease-free survival (DFS) in patients with stage I-III gastric cancer (GC), and its prognostic value is similar to that of the prognostic nutritional index (PNI).

Summary: This study tested the effects of antipsychotic drugs amisulpride and/or aripiprazole on the Sonic hedgehog (Shh)-pathway, cognitive functions, and neurogenesis in a rat model of schizophrenia. The results showed that social isolation led to cognitive dysfunction, disorganized Shh-pathway proteins, and increased glial fibrillary acidic protein (GFAP)-stained astrocytes. However, treatment with these drugs reversed these changes, increasing Shh, transmembrane patched-1 and smoothened, glioma-associated-oncogene (GLI)-1 levels, dopamine-1 receptors and brain derived neurotrophic factor, and decreasing GLI-3 protein, GFAP immune reaction in astrocytes, and inflammatory markers.

Summary: ALDOC is overexpressed in gastric cancer, promoting malignant behavior and serving as an independent risk factor for poor prognosis. miR-19a-5p upregulates ALDOC by downregulating ETS1, leading to poor prognosis. ALDOC is associated with immune infiltration, tumor mutations, and a prognostic model can be established based on ALDOC.

Summary: Piperonyl butoxide (PBO) is a widely used semisynthetic chemical in pesticide formulations, acting as a pesticide synergist and inhibitor of Sonic hedgehog signaling. Animal studies suggest that prenatal exposure to PBO can lead to craniofacial malformations or neurodevelopmental abnormalities, with human studies linking PBO exposure to neurodevelopmental deficits. Further research is needed to better understand the developmental toxicity and potential contribution of PBO to human birth defects.

Summary: The study found up-regulation of SHH and accumulation of SQSTM1/P62 in epithelial ovarian cancer. Cyp induced autophagy through the PI3K/AKT signaling pathway, and low-dose Cyp and chloroquine (CQ) significantly promoted the migratory ability of SKOV3 cells. The findings suggest that inhibition of the SHH pathway and autophagy may be a potential and effective therapy for the treatment of ovarian cancer.

Summary: The study found that apatinib effectively suppressed GCSC traits, including inhibiting tumorsphere formation and cell proliferation, reducing GCSC markers expression and CD133(+) cell number, and inducing apoptosis. Additionally, apatinib downregulated the SHH pathway activation, which was crucial for inhibiting GCSCs.
