Plankton composition and environmental factors contribute to Vibrio seasonality

Plankton represent a nutrient-rich reservoir capable of enriching Vibrio species, which can include human pathogens, at higher densities than the surrounding water column. To better understand the relationship between vibrios and plankton, the partitioning of culturable vibrios, on TCBS, between free living and plankton associated (63-200- and >200-mu m-size fractions) was monitored over a 1-year period in coastal waters of Georgia, USA. Seasonal changes in the total Vibrio concentration were then compared with changes in environmental parameters as well as changes in the relative composition of the plankton community. Using univariate analyses, Vibrio concentrations were strongly associated with temperature, especially when those vibrios were plankton associated (R-2 = 0.69 and 0.88 for the water and both plankton fractions; respectively) (P<0.01). Multivariate general linear models revealed that Vibrio concentrations in the plankton fractions were also correlated to shifts in the relative abundance of specific plankton taxa. In the 63-200-mu m fraction, Vibrio concentrations were inversely associated with copepods, cyanobacteria and diatoms. In the >200-mu m fraction, Vibrio concentrations were positively associated with copepods and negatively associated with decapod larvae. Our results confirm the role of temperature in Vibrio seasonality and highlight an important and independent role for plankton composition in explaining seasonal changes in Vibrio concentration. Building-level wastewater surveillance using tampon swabs and RT-LAMP for rapid SARS-CoV-2 RNA detection

Summary: Wastewater surveillance for SARS-CoV-2 RNA has shown correlation with COVID-19 cases and clinical testing. This study trialed tampons and RT-LAMP for wastewater monitoring, providing rapid results within three hours. Despite lower sensitivity than RT-ddPCR, this method demonstrated comparable predictive values to RT-qPCR and offers a cost-effective and rapid approach for scalable COVID-19 wastewater surveillance.

Non-point source fecal contamination from aging wastewater infrastructure is a primary driver of antibiotic resistance in surface waters

Summary: Antibiotic resistance poses a global threat to human health. This study examined the presence of fecal marker genes and antibiotic resistant genes (ARGs) in stream water samples from the Upper Oconee watershed in Georgia, USA. The results showed widespread fecal contamination from humans, ruminants, and poultry, with a high percentage of samples testing positive for at least one of the six targeted ARGs. Non-point sources of fecal contamination, particularly aging wastewater infrastructure, were identified as critical disseminators of ARGs in the environment.

Use and Evaluation of a pES213-Derived Plasmid for the Constitutive Expression of gfp Protein in Pathogenic Vibrios: a Tagging Tool for In Vitro Studies

Summary: This study demonstrates and evaluates the effectiveness of pVSV102 plasmid as a tool for tagging Vibrio and related species. The results show that pVSV102 is a stable GFP plasmid that can be used to tag a broad range of vibrios and enable easy differentiation within complex mixtures of cells.

Coral Disease and Ingestion: Investigating the Role of Heterotrophy in the Transmission of Pathogenic Vibrio spp. using a Sea Anemone (Exaiptasia pallida) Model System

Summary: The mucus membrane is the front line of defense against pathogens in corals. Understanding disease transmission in corals is complicated, but most of the established transmission pathways are associated with damage rather than evasion of immune defenses. However, this study suggests that ingestion may be an important pathway for the transmission of coral pathogens that bypasses the mucus membrane.
