Paraoerskovia marina gen. nov., sp nov., an actinobacterium isolated from marine sediment

A Gram-positive-staining, facultatively anaerobic bacterial strain, CTT-37(T), was isolated from a marine sediment sample collected from Tottori city, located on the shore of the Sea of Japan. A 16S rRNA gene sequence comparison indicated that the isolate represents a novel clade that clusters with members of the families Cellulomonadaceae and Sanguibacteraceae. Strain CTT-37(T) shared maximum 16S rRNA gene sequence similarity of 96.4% with Oerskovia paurometabola DSM 14281(T) and 96.2% with Oerskovia enterophila DSM 43852(T). The DNA-DNA hybridization value between strain CTT-37(T) and O. enterophila JCM 7350(T) was 10-12%. The following chemotaxonomic characteristics of strain CTT-37(T) were markedly different from those of strains in the genus Oerskovia. The cell wall contained L-serine in the peptidoglycan interpeptide bridge. The predominant menaquinone was MK-9 (H-4); other quinones detected were MK-9 and MK-9(H-2). The only polar lipid was phosphatidylglycerol and the G + C content of the DNA was 70 mol%. Differences in phenotypic characteristics and large phylogenetic distances between strain CTT-37(T) and all members of the genus Oerskovia supported the classification of CTT-37(T) within a new genus and species, for which the name Paraoerskovia marina gen. nov., sp. nov. is proposed. The type strain of Paraoerskovia marina is CTT-37(T) (=NBRC 104352(T)=DSM 21750(T)).

Dunaimycin C3, a new GRP78 downregulator fromStreptomycessp. RAN389

Illumina sequencing of 16S rRNA genes reveals a unique microbial community in three anaerobic sludge digesters of Dubai

Summary: This study analyzed the microbial communities in three anaerobic digesters at the Jebel Ali Sewage Water Treatment Plant in Dubai, United Arab Emirates. The results showed that the bacterial communities were dominated by fermentative and thermophilic bacteria, while the Archaea were mainly Methanocorpusculum and other hydrogenotrophic archaea. The diverse microbial composition in the digesters may be influenced by the climatic conditions and the use of desalinated seawater in Dubai.

Nitrite as a causal factor for nitrate-dependent anaerobic corrosion of metallic iron induced by Prolixibacter strains

Summary: The study isolated four Prolixibacter strains, belonging to the Bacteroidetes phylum, that were able to reduce nitrate under anaerobic conditions, leading to enhanced corrosion of Fe-0 and carbon steel. The corrosion was mainly attributed to biological reduction of nitrate to nitrite followed by chemical oxidation of Fe-0 to Fe2+ and Fe3+.

Additionally, we successfully altered the substrate specificity by introducing a W210A substitution, resulting in a novel farnesyl pyrophosphate C6-methyltransferase.

Summary: In this study, the de novo synthesis of triacylglycerol (TAG) in Coccomyxa sp. Obi strain under nitrogen (N)-replete and N-limiting conditions was analyzed. The results showed that TAG synthesis is limited by fatty acid supply under N-replete conditions, while it is enhanced under N-limiting conditions. The expression of the acyl-ACP thioesterase, FAT1, involved in releasing fatty acids from acyl-ACP, was induced under N-limiting conditions. Furthermore, the turnover of TAG was significant under N-replete conditions. The slow conversion of diacylglycerol to TAG partly explained the slower rate of TAG synthesis compared to polar lipid synthesis under N-limiting conditions.

Acyltransferase Domain Exchange between Two Independent Type I Polyketide Synthases in the Same Producer Strain of Macrolide Antibiotics

Summary: In this study, the biosynthetic gene cluster for virustomycin A, an antibiotic produced by Streptomyces graminofaciens A-8890, was identified. The study also demonstrated the exchangeability of the acyltransferase domain between the PKSs involved in FD-891 biosynthesis and the Vsm PKSs. By exchanging the malonyltransferase domain in the loading module of the PKSs, a two-carbon-elongated analog 26-ethyl-FD-891 was successfully produced with potent cytotoxic activity.

Mechanism of S-Adenosyl-L-methionine C-Methylation by Cobalamin-dependent Radical S-Adenosyl-L-methionine Methylase in 1-Amino-2-methylcyclopropanecarboxylic Acid Biosynthesis

Summary: This study determined that the radical SAM methylase Orf29 catalyzes the C-methylation of SAM to produce (4'' R)-4''-methyl-SAM. It was also found that the 5'-deoxyadenosyl radical generated by Orf29 extracts the pro-R hydrogen atom from the C-4'' position of SAM to generate a radical intermediate, which reacts with methylcobalamin to form (4'' R)-4''-methyl-SAM. Therefore, the Orf29-catalyzed C-methylation reaction was confirmed to proceed with retention of configuration.

Notch activator cyclopiazonic acid induces apoptosis in HL-60 cells through calcineurin activation

Summary: We screened a library of microbial extracts and compounds and identified Pulicatins F and G, as well as cyclopiazonic acid (CPA), as Notch activators. The synthesized Pulicatin F and G showed activation activity, while CPA increased protein levels and mRNA expression of HES1. CPA also inhibited the expression of FLT3, which is known to inhibit apoptosis, and induced apoptosis in HL-60 cells. The Notch activation by CPA was canceled by the addition of FK506, an inhibitor of calcineurin.
