Gene expression following ionising radiation: Identification of biomarkers for dose estimation and prediction of individual response

Materials and methods: aEuro integral Analysis of gene expression in response to radiation was carried out using three independent techniques (Microarray, Multiplex Quantitative Real-Time Polymerase Chain Reaction (MQRT- PCR) and nCounter (R) A (R) Analysis System) in human dividing lymphocytes in culture and peripheral blood leukocytes exposed ex vivo from the same donors. Results: aEuro integral Variations in transcriptional response to exposure to ionising radiation analysed by microarray allowed the identification of genes which can be measured accurately using MQRT PCR and another technique allowing direct count of mRNA copies. We have identified genes which are consistently up-regulated following exposure to 2 or 4 Gy of X-rays at different time points, for all individuals in blood and cultured lymphocytes. Down-regulated genes including cyclins, centromeric and mitotic checkpoint genes, particularly those associated with chromosome instability and cancer could be detected in dividing lymphocytes only. Conclusions: aEuro integral The data provide evidence that there are a number of genes which seem suitable for biological dosimetry using peripheral blood, including sestrin 1 (SESN1), growth arrest and DNA damage inducible 45 alpha (GADD45A), cyclin-dependent kinase inhibitor 1A (CDKN1A), cyclin G1 (CCNG1), ferredoxin reductase (FDXR), p53 up-regulated mediator of apoptosis (BBC3) and Mdm2 p53 binding protein homolog (MDM2). These biomarkers could potentially be used for triage after large-scale radiological incidents and for monitoring radiation exposure during radiotherapy.

Summary: AML is an aggressive cancer with rapid progression, where cytogenetic and genetic analysis play crucial roles in diagnosis and prognosis. This study identified two novel deleterious mutations in DNMT3A and RUNX1 genes, providing additional information for patient diagnosis and prognosis. The exclusion and co-occurrence of specific gene mutation patterns in AML were also confirmed, offering insights into the mechanisms of AML leukaemogenesis.

Modeling low-dose radiation-induced acute myeloid leukemia in male CBA/H mice

Summary: Hypothermic conditions following irradiation significantly inhibited transcriptional responses to ionizing radiation exposure, but sample shipment at different temperatures did not affect gene expression level except for SESN1. The transcriptional response to IR of specific genes depended on the cell fraction used, while FDXR and CCNG1 showed consistent response across all conditions tested.

Additionally, the upregulation of FDXR gene transcription was dose-dependent and could provide individual biological dosimetry information for predicting radiotherapy-associated adverse health outcomes.

Summary: Extracellular vesicles (EVs) play an important role in mediating bystander responses in irradiated bone marrow by altering cellular pathways through the miRNAs they carry. Using a mouse model, we analyzed the miRNA content of BM-derived EVs from mice irradiated with different doses and investigated the proteomic changes in BM cells upon direct irradiation or treatment with EVs. We identified key cellular processes regulated by miRNAs and suggested potential candidates involved in the regulation of these bystander processes.

Can We Detect T Cell Receptors from Long-Read RNA-Seq Data?

Summary: This study aimed to investigate the feasibility of utilizing existing methods for TR sequence annotation in long-read sequencing data. TRUST4 algorithm detected the largest number of CDR3 sequences, MiXCR and TRUST4 algorithms showed the same distribution in annotated V and J genes, which can be used to analyze the repertoire of V/J genes used in rearranged TR genes.
