A new highly viscoelastic hyaluronic acid gel: rheological properties, biocompatibility and clinical investigation in esthetic and restorative surgery

Nowadays there is an increased demand for safe and effective volume enhancing fillers to achieve soft tissue augmentation in order to overcome tissue defects and aging-associated skin changes. In the present study we characterized the rheological and biological properties of Variofill (R), a new highly viscoelastic hyaluronic acid gel, by investigating the local effects following subcutaneous implantation in the rat to detect the host-tissue reactions and biodegradation over 18 months. We also investigated, for the first time, the application of Variofill (R) in esthetic and restorative surgery in two medical case reports. In the first case report we successfully performed Variofill (R) treatment to improve facial scars in a patient previously involved in a car crash. In the second case report we carried out a novel procedure involving a high-dose (1000 ml) injection of Variofill (R) into the dermis and subcutis of the abdominal quadrants in order to allow a classic reconstructive procedure of the abdominal wall in a patient presenting a wide incisional hernia. Current diagnosis relies on clinical symptoms as there are no specific diagnostic markers available, and more definite treatment strategies are required. Research on the pathophysiology of MCS, particularly focusing on brain networks, environmental pollution, and correlation with other pathologies, is ongoing to better understand and develop treatment strategies.

Evidence for Endogenous Opioid Dependence Related to Latent Sensitization in a Rat Model of Chronic Inflammatory Pain

Summary: Studies in mice have shown that constitutive activation of endogenous opioid signaling leads to both endogenous analgesia and opioid dependence. This study aimed to determine if a similar pattern exists in rats. Rats with chronic inflammatory pain were treated with different doses of the opioid receptor inverse agonist naltrexone (NTX) and monitored for withdrawal behaviors. The response to NTX in the rat model was different from the mouse model, suggesting species-related differences in pharmacological response and highlighting the importance of species selection in preclinical pain research.

3D human foreskin model for testing topical formulations of sildenafil citrate

Summary: This study aimed to investigate the loading of sildenafil citrate in three commercial transdermal vehicles using 3D full-thickness skin equivalent and compare the results with permeability experiments using porcine skin. The results showed that the results obtained using the 3D skin equivalent were comparable to those obtained using porcine skin, suggesting that the 3D skin model can be a valid alternative for ex-vivo skin absorption experiments.
