Nanotoxicity Overview: Nano-Threat to Susceptible Populations

Due to the increasing applications of nanomaterials and nanotechnology, potential danger of nanoparticle exposure has become a critical issue. However, recent nanotoxicity studies have mainly focused on the health risks to healthy adult population. The nanotoxicity effects on susceptible populations (such as pregnant, neonate, diseased, and aged populations) have been overlooked. Due to the alterations in physiological structures and functions in susceptible populations, they often suffer more damage from the same exposure. Thus, it is urgent to understand the effects of nanoparticle exposure on these populations. In order to fill this gap, the potential effects of nanoparticles to pregnant females, neonate, diseased, and aged population, as well as the possible underlying mechanisms are reviewed in this article. Investigations show that responses from susceptible population to nanoparticle exposure are often more severe. Reduced protection mechanism, compromised immunity, and impaired self-repair ability in these susceptible populations may contribute to the aggravated toxicity effects. This review will help minimize adverse effects of nanoparticles to susceptible population in future nanotechnology applications.

Paper Unraveling the joint toxicity of transition-metal dichalcogenides and per- and polyfluoroalkyl substances in aqueous mediums by experimentation, machine learning and molecular dynamics

Summary: The interactions between transition-metal dichalcogenides (TMDCs) and per- and polyfluoroalkyl substances (PFAS) were explored using experimental methods and machine learning approaches. The study revealed that TMDCs-PFAS interactions have unique toxicity effects on cell lines and enhance the bioaccumulation of TMDCs in zebrafish, leading to oxidative stress and histopathological alterations. This research emphasizes the importance of investigating interactions between emerging and existing contaminants in aquatic environments using state-of-art techniques to safeguard environmental health.

Zn2+ loading as a critical contributor to the circ_0008553-mediated oxidative stress and inflammation in response to PM2.5 exposures

Summary: Inflammation is a major adverse outcome induced by PM2.5, and plays a critical role in PM2.5 exposure-associated diseases. This study identified a novel circRNA, circ_0008553, that is responsible for the PM2.5-activated inflammatory response in human bronchial epithelial cells. The findings highlight the significance of this circRNA in the intervention of PM2.5-induced adverse outcomes.

Nonmonotonic effect of CuO nanoparticles on medium-chain carboxylates production from waste activated sludge

Summary: The applications of CuO nanoparticles in industrial and agriculture have led to their increased concentrations in wastewater and accumulation in waste activated sludge, raising concerns about their impact on the production of medium-chain carboxylates (MCCs) from anaerobic fermentation of sludge. Our study showed that CuO NPs at 10-50 mg/g-TS significantly inhibit MCCs production, with reactive oxygen species generation being the key factor. However, at lower CuO NPs concentrations (0.5-2.5 mg/g-TS), MCCs production was enhanced. Molecular approaches and metaproteomic analysis revealed the impact of CuO NPs on microbial processes and protein expression levels related to MCCs production.

Oxidized Graphene Alleviates Benzo[a]pyrene-Induced Cytotoxicity by Suppressing Mitochondrial Apoptosis Pathways

Summary: Environmental carbon nanomaterials, such as multi-walled carbon nanotubes and graphene oxides, interact with aromatic hydrocarbon pollutants, resulting in adverse effects on ecosystems and human health. The combined toxicity of carbon nanomaterials and benzo[a]pyrene is still unknown. Among the investigated carbon nanomaterials, MWCNTs-NH2, p-GO, and GO-COOH showed some alleviation of the cytotoxicity induced by benzo[a]pyrene, with GO-COOH exhibiting the strongest inhibitory effect. Mechanistically, GO-COOH reduced benzo[a]pyrene-induced cell apoptosis and cytotoxicity by regulating apoptotic proteins and maintaining mitochondrial membrane potential. These findings highlight the importance of the physicochemical properties of carbon nanomaterials in modulating the cytotoxicity of benzo[a]pyrene.

Computer-Aided Discovery of Potent Broad-Spectrum Vaccine Adjuvants

Summary: Using computer-aided molecule design and machine learning, researchers have discovered two new broad-spectrum adjuvants that can enhance vaccine responses. These adjuvants can activate dendritic cells through multiple toll-like receptors, promote antigen presentation to T cells, and elicit strong immune responses in mice, effectively suppressing tumor growth and metastasis.

Chemical speciation determines combined cytotoxicity: Examples of biochar and arsenic/chromium

Summary: Biochars (BCs) can interact with multivalent metal ions in the environment, causing changes in their ionic valence states and resulting in unknown combined toxicity. In this study, we investigated the interaction between BCs and Cr or As and their combined cytotoxicity in human colorectal mucosal cells. Our results showed that the redox-induced valence state change is a critical factor in the combined cytotoxicity of BCs with Cr/As.

Regulating the reaction pathway of nZVI to improve the decontamination performance through magnetic spatial confinement effect

Summary: This study investigates the effect of constructing a magnetic mesoporous SiO2 shell on the surface of nanoscale zero-valent iron (nZVI). The results show that this structure can regulate the electron transfer pattern, avoid the formation of iron oxide layers, and improve the efficiency of active hydrogen production.

Sequential exposures of single walled carbon nanotubes and heavy metal ions to macrophages induce different cytotoxicity

Summary: The probability of occupational exposure to carbon nanotubes (CNTs) is increasing due to their growing production and biomedical applications. As a result, the risk of co-exposure of nanomaterials with environmental pollutants is also increasing. This study investigated the toxicity of pristine single-walled CNTs (p-SWCNTs) and oxidized single-walled CNTs (SWCNT-COOHs) on cell responses to metal ions (Pb2+, Cu2+, and Cr(VI)). The findings suggest that understanding the effects of the exposure sequence of engineered nanomaterials and environmental pollutants on their toxicity is crucial for evaluating combined toxicity.

ILTox: A Curated Toxicity Database for Machine Learning and Design of Environmentally Friendly Ionic Liquids

Summary: A comprehensive online database on the toxicity of ionic liquids (ILs) has been developed to aid in the design of environmentally friendly ILs using machine learning. The ILTox database contains manually curated data, including 1183 ILs and over 6700 pieces of toxicity data across different organisms. Machine learning models have been constructed using this database to analyze the relationship between IL structures and toxicities. The database has been used for virtual screening of desired IL properties from a large pool of ILs and has proven to be a valuable resource in understanding IL toxicity.

SERS for Probing Highly Reactive Pd Sites with Elongated Pd-Pd Bonds on the Au Surface

Summary: Bond length is an important structural parameter that can affect the activity of catalysts. In core-shell gold-palladium (Au@Pd) nanocatalysts, elongated Pd-Pd bonds resulting from lattice mismatches between Au and Pd are found to exhibit distinct catalytic activity. Surface-enhanced Raman scattering (SERS) spectra of chemisorbed 2,6-dimethylphenyl isocyanide (2,6-DMPI) were used to investigate the elongated Pd-Pd bonds on Au@Pd. The study revealed that strained sites with elongated Pd bonds are more than 100 times more active in a catalyzed Suzuki-Miyaura coupling reaction.

Charged nanoplastics differentially affect the conjugative transfer of antibiotic resistance genes

Summary: This study investigated the impact of nanoplastics (NPs) on the spread of antibiotic resistance genes (ARGs) and found that negatively charged polystyrene (PS) and PS-COOH enhanced ARG transfer at low concentrations, while transfer declined at higher concentrations. Conversely, PS-NH2 increased ARG transfer at all tested concentrations, with the highest promotion effect at 500 mg L-1. The effects on ARG transfer were a combination of ROS production-induced promotion and NP agglomeration-caused inhibition at high concentrations.

Inflammatory responses induced by synergistic actions between nanoplastics and typical heavy metal ions in human cells

Summary: Nanoplastics can transport heavy metal ions and pose health risks when ingested through the food chain. This study investigated the induction of inflammatory responses in human cells by ion-adsorbed polystyrene NPs and found that intracellular ions played a role in the inflammation caused by these NPs. Machine learning models identified Cd2+ and Pb2+ as crucial for the increased expression of pro-inflammatory cytokines in different cell types. The activation of NF-kappa B and ERK1/2 pathways were associated with the high expression of pro-inflammatory cytokines in the respective cell types.

The role of crystallinity of palladium nanocrystals in ROS generation and cytotoxicity induction

Summary: In this study, palladium (Pd) nanocrystals with different crystalline forms were synthesized, and the underlying mechanisms of ROS-mediated cytotoxicity were revealed. Nanocubes and nanooctahedrons/nanotetrahedrons triggered cytotoxicity by generating singlet oxygen (O-1(2)) and hydroxyl radicals (OH), respectively. Nanoconcave-tetrahedrons induced ROS-mediated cytotoxicity via activating the superoxide (O-2(-)) pathway. The alkalization of pH resulting from intracellular ROS conversion was identified as a new biomarker for analyzing cytotoxicity.

A bioinspired silica nanocomposite for enhanced multidrug-resistant bacteria treatment and wash-free imaging

Summary: In this study, a novel antibacterial composite nanomaterial based on spiky mesoporous silica spheres loaded with poly(ionic liquid)s and aggregation-induced emission luminogens (AIEgens) was designed for efficient treatment and imaging of multidrug-resistant (MDR) bacteria. The nanocomposite exhibited excellent and long-term antibacterial activity towards both Gram-negative and Gram-positive bacteria. Additionally, fluorescent AIEgens facilitated real-time bacterial imaging. This study provides a multifunctional platform and a promising alternative to antibiotics for combating pathogenic MDR bacteria.
