Summary: Biotransformation of flavonoid dimer FD18 resulted in an active metabolite FM04, which exhibited improved druggability and was more potent in reversing P-gp-mediated PTX resistance. FM04 sensitized cancer cells to multiple anticancer drugs by inhibiting P-gp transport activity and stimulating P-gp ATPase. In addition, FM04 enhanced the intestinal absorption of PTX and showed significant antitumor activity in mouse models.

Summary: This study found that certain flavonoids can effectively reverse multidrug resistance mediated by P-glycoprotein (P-gp) by inhibiting its activity, with specific structural groups and substitutions playing a crucial role in their inhibitory effects. Additionally, flavonoids can impact P-gp ATPase activity and expression levels, thereby influencing its drug transport activity.

Summary: Traditional methodologies for assessing chemical toxicity are expensive and time-consuming, but computational modeling approaches, particularly QSAR models, provide a low-cost alternative. However, conventional QSAR models have limited training data and poor predictivity for new compounds. This study developed data-driven models for carcinogenicity and successfully identified potential new human carcinogens.

Summary: This study found that acetylation could significantly enhance the anticancer activity and oral bioavailability of 5-DTAN. 5-ATAN showed stronger cytotoxic effects and cell cycle arrest in PC-3 prostate cancer cells compared to TAN and 5-DTAN. It induced apoptosis, inhibited cell migration, and had a more extensive distribution in mice, indicating its potential as a strategy to enhance the bioactivities of natural products.

Summary: Panax ginseng Meyer is a traditional Chinese medicine widely used as a tonic in Asia. The main active components, dammarane-type ginsenosides, have shown various pharmacological activities, but their oral bioavailability is limited, restricting their clinical use. Hence, improving the oral bioavailability of ginsenosides is crucial.

Summary: This study reports a case of an intellectual disability patient who developed opioid use disorder after using low to moderate doses of loperamide. The patient had reduced enzyme activity, leading to enhanced effects of loperamide on the central nervous system and contributing to the development of substance use disorder. The use of slow release oral morphine effectively controlled loperamide use and improved the patient's clinical condition. This case highlights the need for caution when prescribing loperamide, especially in vulnerable individuals with limited cognitive resources and understanding of medication risks.

Summary: This article proposes a hybrid contextual semantic network (HCSNet) that accurately and simultaneously segments and detects small-size stroke lesions from magnetic resonance images. HCSNet incorporates an encoder-decoder architecture and a novel hybrid contextual semantic module to generate high-quality contextual semantic features. A mixing-loss function is also proposed to optimize HCSNet for unbalanced small-size lesions. Extensive experiments on 2D magnetic resonance images from the ATLAS R2.0 dataset demonstrate that HCSNet outperforms state-of-the-art methods in segmenting and detecting small-size stroke lesions. Visualization and ablation experiments further confirm the improved performance of HCSNet with the hybrid semantic module.
