Summary: In this study, the role of the Slit-Robo3 signaling pathway in the morphogenesis of the Drosophila optic lobe was investigated. It was found that Slit and Robo3 are expressed in the same neuronal population and they regulate the compartmentalization of the optic lobe through an autocrine/paracrine mechanism. Furthermore, interactions with genes involved in endocytosis and actin dynamics were detected, indicating the involvement of the endocytic pathway and small GTPases in this process. This study reveals the important role of the Slit-Robo3 pathway in CNS development.

Understanding the complexity of circuits requires identifying unknown factors and dissecting molecular topographies. Glial cells contribute to axon guidance, but their molecular mechanisms are not fully understood. Studying the roles of glia in different models can expand our understanding of their functions.

Summary: Axon degeneration and NMJ disruption are crucial in ALS, but the mechanisms leading to these pathologies remain unclear. TDP-43 mislocalization and accumulation are common in ALS patients and have been observed in intramuscular nerves of early symptomatic patients. This suggests a gain of function for TDP-43 in axons, potentially contributing to early NMJ disruption. This review discusses the process of TDP-43 accumulation and phosphorylation in axons, its role in healthy axons, and possible mechanisms connecting TDP-43 pathology to axon and NMJ degeneration in ALS.

Summary: Mutations in UBQLN2 cause X-linked forms of ALS and FTD, but the underlying mechanisms are unknown. Studying ALS-associated mutants of UBQLN2 in Drosophila, it was found that they trigger heat stress-dependent neurodegeneration. A genetic modifier screen identified endolysosomal and axon guidance genes, including the netrin receptor Unc-5, as key regulators of UBQLN2 toxicity. Reduced dosage of Unc-5 or its coreceptor Dcc/frazzled alleviated neurodegenerative phenotypes in flies expressing UBQLN2(ALS) alleles. Further experiments using iPSCs and iMNs demonstrated lysosomal defects and cytosolic UBQLN2 inclusions, neurite complexity reduction, and growth cone defects, which were partially reversed by silencing of UNC5B and DCC. These findings suggest altered growth cone dynamics as a conserved pathomechanism in UBQLN2-associated ALS/FTD.
