However, this approach is ineffective for BRAF-WT melanoma, and relapse often occurs after initial regression. Inhibiting MAP kinase pathways downstream at ERK1/2 or targeting antiapoptotic Bcl-2 proteins like Mcl-1 may provide alternative strategies.

Summary: The pH-sensitive nanoparticles D/D NPs demonstrated significant anticancer activity in breast cancer cells by enhancing apoptosis and activating mitochondrial apoptosis pathway. In vivo studies showed that D/D NPs effectively inhibited tumor growth and prevented lung metastasis in a breast cancer mouse model, without causing distinct changes in major organs. These results suggest that D/D NP-based combination therapy may be a promising strategy for the treatment of metastatic breast cancers through ROS-mediated mitochondrial apoptosis pathway.

Summary: Dysregulation of mitochondrial calcium uniporter (MCU) plays a carcinogenic role in several cancers, including oral squamous cell carcinoma (OSCC). The study aimed to investigate the role of MCU in OSCC and its regulatory relationship with dihydroartemisinin (DHA). The results showed that MCU and its complex members were overexpressed in OSCC and DHA treatment suppressed MCU expression. MCU overexpression reversed the inhibitory effects of DHA on other proteins. Knockdown of MCU or DHA treatment suppressed proliferation and migration of OSCC cells.

This study demonstrates that AC can induce both reactive oxygen species (ROS)-dependent apoptosis and ferroptosis in tumor cells. The Bim- and Bax-mediated mitochondrial pathways play a dominant role in AC-induced ferroptosis and apoptosis.

Summary: The level of mitochondrial apoptotic priming is a critical determinant of cell fate upon p53 reactivation, with cells having high initial priming tending to undergo apoptosis and cells with low priming tending to survive and arrest in the cell cycle. Manipulating the priming levels through BCL-2 or BCL-XL expression or inhibition can affect the outcome of p53 restoration, highlighting mitochondrial apoptotic priming as a key factor in determining cell fate. Moreover, less primed cells can be forced into apoptotic cell fate following p53 activation using p53-independent drugs that increase apoptotic priming.
