Objective Copy number variants (CNVs) have been associated with the risk to develop multiple autoimmune diseases. Our objective was to identify CNVs associated with the risk to develop psoriatic arthritis (PsA) using a genome-wide analysis approach. Methods A total of 835 patients with PsA and 1498 healthy controls were genotyped for CNVs using the Illumina HumanHap610 BeadChip genotyping platform. Genomic CNVs were characterised using CNstream analysis software and analysed for association using the chi(2) test. The most significant genomic CNV associations with PsA risk were independently tested in a validation sample of 1133 patients with PsA and 1831 healthy controls. In order to test for the specificity of the variants with PsA aetiology, we also analysed the association to a cohort of 822 patients with purely cutaneous psoriasis (PsC). Results A total of 165 common CNVs were identified in the genome-wide analysis. We found a highly significant association of an intergenic deletion between ADAMTS9 and MAGI1 genes on chromosome 3p14.1 (p=0.00014). Using the independent patient and control cohort, we validated the association between ADAMTS9-MAGI1 deletion and PsA risk (p=0.032). Using next-generation sequencing, we characterised the 26 kb associated deletion. Finally, analysing the PsC cohort we found a lower frequency of the deletion compared with the PsA cohort (p=0.0088) and a similar frequency to that of healthy controls (p>0.3). Conclusions The present genome-wide scan for CNVs associated with PsA risk has identified a new deletion associated with disease risk and which is also differential from PsC risk.

Strategies and resources to optimise the management of Psoriatic Arthritis patients: the CREA Project

Raquel Almodovara, Juan D. Canete, Eduardo Collantes, Eugenio de Miguel, Cristina Fernandez Carballido, Jordi Gratacos, Xavier Juanola, Jose A. Pinto, Ruben Queiro, Pedro Zarco

Summary: The aim of the study was to propose strategies to improve the initial assessment and follow-up of patients with psoriatic arthritis (PsA) in Spain. A survey among expert rheumatologists in Spain was conducted, and a total of 108 strategies were proposed, including assessment of quality of life and disease-impact indices, comorbidities and extra-articular manifestations, laboratory tests, imaging tests, etc. Overall, these strategies are applicable to the entire country and are of significant importance in meeting the needs of PsA patients and ensuring optimal follow-up.

Summary: This article summarizes existing evidence regarding pharmaceutical and nonpharmaceutical interventions for enthesitis in patients with psoriatic arthritis (PsA) to facilitate an evidence-based update of the Group for Research and Assessment in Psoriasis and Psoriatic Arthritis (GRAPPA) treatment recommendations for PsA. The article found several classes of systemic conventional and advanced therapies and local measures were recommended for active enthesitis in patients with PsA.

Summary: The aim of this study was to investigate the role of endothelin-1 (ET-1) as a biomarker of interstitial lung disease (ILD), and to differentiate between idiopathic pulmonary fibrosis (IPF) and ILD associated with autoimmune diseases (AD-ILD). The study included a large cohort of ILD patients and healthy controls, and found that ET-1 levels were increased in IPF and AD-ILD patients compared to controls. However, ET-1 levels could not differentiate between IPF and AD-ILD, and were associated with worse lung function in IPF and RA-ILD patients.

Summary: This study compares the efficacy of tumor necrosis factor inhibitors (adalimumab and infliximab) with tocilizumab in patients with refractory cystoid macular edema (CME) due to Behcet's disease (BD). The conclusion is that both adalimumab, infliximab, and tocilizumab can effectively treat refractory CME associated with BD uveitis.

Summary: This study investigates the role of serum ANGPTL2 levels in Hidradenitis suppurativa (HS) patients and controls. It is found that HS patients have higher serum ANGPTL2 levels compared to controls, and these levels are positively correlated with disease duration and severity. Therefore, ANGPTL2 may serve as a biomarker for HS severity.
