Impact of insufficient sleep on dysregulated blood glucose control under standardised meal conditions

Neli Tsereteli, Raphael Vallat, Juan Fernandez-Tajes, Linda M. Delahanty, Jose M. Ordovas, David A. Drew, Ana M. Valdes, Nicola Segata, Andrew T. Chan, Jonathan Wolf, Sarah E. Berry, Matthew P. Walker, Timothy D. Spector, Paul W. Franks

Summary: Sleep quality, duration, and timing have significant associations with postprandial glycemic responses, with poor sleep efficiency and later bedtime routines leading to more pronounced blood glucose levels after breakfast. Deviation from usual sleep patterns also affects postprandial glycemic control, highlighting the importance of sleep as a modifiable factor in optimizing metabolic health.

Summary: SARS-CoV-2 infection can lead to a range of outcomes, influenced by factors such as challenge dose, age, gender, comorbidity, and host genomic variation. Human leukocyte antigen (HLA) polymorphisms may affect immune response and disease outcomes. This study investigated the association between HLA-II alleles and symptomatic COVID-19, virus-specific antibody, and T-cell immunity in UK healthcare workers (HCWs). The results showed that certain HLA-II alleles were associated with an increased risk of symptomatic COVID-19 and altered immune responsiveness to viral proteins.

Upregulated expression of FFAR2 and SOC3 genes is associated with gout

Oliver C. Orji, Maria B. Lopez-Dominguez, Gabriela Sandoval-Plata, Tamar Guetta-Baranes, Ana M. Valdes, Michael Doherty, Kevin Morgan, Abhishek Abhishek

Summary: This study found that the expression of FFAR2 and SOCS3 genes is significantly upregulated in gout flares and inter-critical gout. FFAR2 may be associated with gout flares, while SOCS3 may play a role in ameliorating gout flares.

Harmonising knee pain patient-reported outcomes: a systematic literature review and meta-analysis of Patient Acceptable Symptom State (PASS) and individual participant data (IPD)

V. Georgopoulos, S. Smith, D. F. McWilliams, M. P. M. Steultjens, A. Williams, A. Price, A. M. Valdes, T. L. Vincent, F. E. Watt, D. A. Walsh

Summary: This study aimed to harmonize patient-reported outcome measures (PROMs) in knee pain due to osteoarthritis or knee trauma using the Patient Acceptable Symptom State scores (PASS) as a criterion. Standardized PROMs showed similar PASS thresholds in group data, but divergent scores within individuals.

Serum Metabolome Analysis Identified Amino-Acid Metabolism Associated With Pain in People With Symptomatic Knee Osteoarthritis-A Cross-Sectional Study

Summary: The study aimed to identify metabolic biomarkers for knee pain and symptomatic osteoarthritis (OA) using metabolic phenotyping. The results showed significant associations between acyl ornithine, carnosine, cortisol, cortisone, cystine, DOPA, glycolithocholic acid sulphate (GLCAS), phenylethylamine (PEA), succinic acid, and pain scores. These metabolites were also found to be associated with inflammatory cytokines.

Cardiometabolic health, diet and the gut microbiome: a meta-omics perspective

Mireia Valles-Colomer, Cristina Menni, Sarah E. Berry, Ana M. Valdes, Tim D. Spector, Nicola Segata

Summary: Cardiometabolic diseases are closely related to the gut microbiome and diet, and high-throughput meta-omics techniques have the potential to reveal their intricate links. However, effective integration of these techniques and their application on large cohorts is still a challenge. This review discusses the potential of meta-omics technologies in improving cardiometabolic health and highlights recent large-scale efforts and insights provided.

Summary: This study characterized the variability and correlations of postprandial metabolite changes in a large cohort. The majority of metabolites showed significant changes after a meal, particularly in very large lipoprotein particles and ketone bodies. Strong correlations were observed between fasting and postprandial metabolites, although some metabolites showed weak correlations.

Combining single-cell RNA sequencing and population-based studies reveals hand osteoarthritis-associated chondrocyte subpopulations and pathways

Hui Li, Xiaofeng Jiang, Yongbing Xiao, Yuqing Zhang, Weiya Zhang, Michael Doherty, Jacquelyn Nestor, Changjun Li, Jing Ye, Tingting Sha, Houchen Lyu, Jie Wei, Chao Zeng, Guanghua Lei

Summary: Hand osteoarthritis is a common joint disorder with unclear molecular mechanisms and lack of disease-modifying drugs. This study used single-cell RNA sequencing analysis to compare the gene expression and cellular composition in osteoarthritic cartilage and nonosteoarthritic cartilage. Inflammatory chondrocytes and fibrocartilage chondrocytes were identified as key subpopulations, and the ferroptosis pathway was found to play a role in hand osteoarthritis. These findings provide new insights into the pathophysiology and potential therapeutic targets for hand osteoarthritis.
