BERBERINE REDUCES LEUKOCYTE ADHESION TO LPS-STIMULATED ENDOTHELIAL CELLS AND VCAM-1 EXPRESSION BOTH IN VIVO AND IN VITRO

Leukocyte adhesion to endothelium plays a critical initiating role in inflammation. Berberine, an anti-inflammatory natural compound, is known to attenuate lipopolysaccharide (LPS)-induced lung injury and improve survival of endotoxemic animals with mechanism not fully clarified. This study investigated the effects of berberine on the LPS-induced leukocyte-endothelial cell adhesion both in vivo and in vitro. We first established an animal model to observe the in vivo LPS-induced adhesion of leukocytes to the endothelium of venules in the lung tissue dose-dependently. Pretreatment of LPS-stimulated rats with berberine for 1 h reduced the leukocyte-endothelium adhesion and vascular cell adhesion molecule-1 (VCAM-1) expression in lung. Pretreatment of LPS-stimulated vascular endothelial cells with berberine also dose-dependently decreased the number of adhered THP-1 cells and VCAM-1 expression at both RNA and protein levels. Berberine was further confirmed to inhibit the nuclear translocation and DNA binding activity of LPS-activated nuclear factor-kappa B (NF-kappa B). These data demonstrated an additional molecular mechanism for the profound anti-inflammatory effect of berberine.

Highly Selective SERCA2a Activators: Preclinical Development of a Congeneric Group of First-in-Class Drug Leads against Heart Failure

Summary: Stimulation of sarcoplasmic reticulum calcium ATPase SERCA2a is a promising therapeutic strategy for heart failure. Istaroxime, a compound with dual mechanisms of action, inhibits Na+/K+ ATPase and stimulates SERCA2a. By synthesizing a series of analogues, we identified a compound that lacks Na+/K+ ATPase inhibitory activity but retains SERCA2a stimulatory action. Further studies in isolated cardiomyocytes and a rat model showed that the compound is safe and improves cardiac performance, making it a potential drug candidate for heart failure treatment.

Derangements and Reversibility of Energy Metabolism in Failing Hearts Resulting from Volume Overload: Transcriptomics and Metabolomics Analyses

Summary: This study investigates the changes and reversibility of cardiac energetics in a rat model of cardiac volume overload (VO) using transcriptomics and metabolomics analyses. The findings suggest that glycolysis is downregulated and redirected towards the hexosamine biosynthetic pathway (HBP) and O-linked-N-acetylglucosaminylation in the compensated hypertrophy (CH) phase, and further suppressed in the progression to heart failure (HF). Derangements in fatty acid oxidation become prominent only in HF. Correction of VO can reverse the differential expression of genes involved in glycolysis, HBP, and fatty acid oxidation in CH but not in HF.

Osteopontin mediation of disturbed flow-induced endothelial mesenchymal transition through CD44 is a novel mechanism of neointimal hyperplasia in arteriovenous fistulae for hemodialysis access

Summary: Our study suggests that disturbed flow in dysfunctional arteriovenous fistulae (AVF) contributes to the development of neointimal hyperplasia (NH) through the induction of endothelial mesenchymal transition (EndMT) via the osteopontin/CD44 axis. We provided evidence from both animal and cell culture experiments to support this hypothesis. We also found that the osteopontin/CD44 axis is involved in AVF dysfunction in patients with uremia. These findings highlight the potential of targeting the osteopontin/CD44 axis as a therapeutic strategy for preventing AVF dysfunction.

High-phosphate diet causes atrial remodeling and increases atrial fibrillation vulnerability via STAT3/NF-κB signaling and oxidative stress

Summary: This study found that high phosphate intake causes atrial remodeling and increases the risk of atrial fibrillation. In mice with chronic kidney disease, a high phosphate diet increased atrial fibrillation vulnerability, atrial fibrosis, and oxidative stress. Inhibiting STAT3/NF-kappa B signaling and oxidative stress can alleviate the atrial abnormalities caused by high phosphate intake. Therefore, high dietary phosphate intake may have detrimental effects on the atria and increase the risk of atrial fibrillation.
