Summary: Viral infections induce systemic cytokinemia. Vaccines can use virus-derived nucleic acids as adjuvants, activating dendritic cells (DCs) through Toll-like receptors (TLRs) in endosomes. Two subsets of DCs, CD141(+)DCs expressing TLR3 and plasmacytoid DCs (pDCs) expressing TLR7/9, play different roles in antiviral immunity. The mode of vaccine sought in infectious diseases and cancer differs based on prophylactic or therapeutic purposes and antigen delivery to cDCs. Adjuvants can be selected on a case-by-case basis.

Summary: The sensitive detection of carcinoembryonic antigen (CEA) was achieved using an electrochemical immunosensor loaded with gold nanoparticles and flower-shaped CuS nanoflowers with peroxidase-like activity. This immunosensor exhibited excellent analytical performance for the quantitative detection of CEA.

Summary: The study aims to develop a new treatment for advanced gastrointestinal cancers. The B7-H6/CD3 ITE showed potent antitumor activity by killing B7-H6-positive tumor cells and activating and proliferating T cells with cytokine secretion. Furthermore, it recruited T cells into tumor tissues and resulted in tumor regression. Further clinical studies are warranted to evaluate the effectiveness of this approach.

Summary: This study aimed to evaluate the diagnostic accuracy of CEA for detecting colorectal cancer recurrence in patients with normal and elevated initial CEA levels. The results showed that the diagnostic accuracy of CEA depends on the initial CEA level, and an increase in CEA level at recurrence is significantly associated with overall survival in patients with recurrent colorectal cancer.

Summary: Blastic plasmacytoid dendritic cell neoplasm (BPDCN) is a rare and aggressive hematologic disease involving the skin and bone marrow. The immunophenotype of BPDCN is characterized by the expression of CD4, CD56, CD123, TCL-1, and CD303. Current treatment for BPDCN is based on high-dose chemotherapy combined with stem cell transplantation, but targeted therapies have shown great promise. This article focuses on the latest advances in genetics and targeted therapies for BPDCN, providing new ideas for its clinical treatment.

Summary: Gallbladder cancer (GBC) is often diagnosed at late stages and lacks effective treatments. This study investigates the potential of using CAR-T cells targeting CEA as a potential immunotherapy for GBC. Results show that CEA is expressed in a high percentage of GBC tumors, and CAR-T cells can recognize and kill GBC cells expressing CEA. The findings support the further development of CEA-specific CAR-T cells for GBC immunotherapy.

Summary: This retrospective study compared the safety and short-term outcomes of endoscopic and laparoscopic resections for gastric GISTs with a diameter of 2-5 cm. The results showed that endoscopic resection had faster recovery, shorter hospital stays, and lower costs compared to laparoscopic resection. The incidence of adverse events was also significantly lower in the endoscopic group.

The scoring system, Zhongshan Tunnel Score, was proposed based on multivariate logistic regression analysis and showed satisfying discriminatory power and goodness-of-fit in internal validation. It could provide clinicians with references for treatment decisions.
