Summary: This study confirmed the inhibitory effect of metformin on prostate cancer cell lines, with mechanisms involving autophagy and cell cycle arrest at G0/G1 phase. Metformin induced AMPK activation, promoted LC3II expression, and down-regulated p62/SQSTM1 expression. Animal experiments demonstrated that metformin reduced tumor volume, upregulated p-AMPK (Thr172) and LC3B, and downregulated Ki-67 levels without abnormal biochemical indicators.

Summary: The study found that lysosome function is dysregulated in PBMCs isolated from COVID-19 patients, and the biomarker LC3B can be used to differentiate between patients with moderate-to-severe disease and those with asymptomatic or mild disease. Patients with an LC3B concentration<5.5 ng/ml will require early hospital admission for oxygen therapy.

Summary: Although the mechanisms of autophagy have been studied, real-time monitoring of this process is still difficult. Researchers developed a biosensor using FRET to observe the priming of the autophagy player LC3B by ATG4B. This biosensor allows for spatial characterization of the priming activity and quantification of autophagy activation. The study also revealed the regulation of the ATG4B-LC3B axis during mitosis.

Summary: The study shows that CGAS can reduce the abundance of micronuclei by promoting autophagy, thus maintaining micronuclei homeostasis. The direct interaction between CGAS and MAP1LC3B is crucial for the autophagic clearance of micronuclei, leading to inhibition of cGAMP production induced by genotoxic stress. This research reveals an important role of CGAS in the selective autophagic clearance of micronuclei.

Summary: The senescence of lens epithelial cells (LECs) is a major factor leading to age-related cataract (ARC). This study aimed to investigate the mechanism of ARC formation and explore the anti-aging effect of metformin (MET) on ARC. The results showed that MET effectively alleviated the senescence of LECs and the formation of ARC probably through inactivation of the AMPK pathway and augmentation of autophagy.

Summary: The expression of SERCA2 in triple negative breast cancer (TNBC) patients is closely related to disease progression, and it also promotes the growth, migration and chemoresistance of breast cancer cells. Through experiments, it is proven that SERCA2 can interact with LC3B, induce cell autophagy, and increase the tolerance of breast cancer cells to drugs. In addition, it is found that targeting SERCA2 can increase cell autophagy and induce cell death, achieving the therapeutic effect of treating breast cancer.

Summary: Autophagy plays a dual role in oncogenesis processes, enhancing cell resistance to oncogenic factors while also participating in tumor progression. This study found associations between the effectiveness of the FLOT regimen in resectable gastric cancers and the key autophagy-related proteins LC3B, mTOR, and AMPK. These proteins were identified as molecular targets affecting cancer progression, metastasis, and the effectiveness of neoadjuvant chemotherapy.

Summary: Autophagic flux, a critical cellular process, can be increased by targeting nutrition-related biochemical signaling. This study introduces a method to directly measure human autophagic flux from blood samples by treating whole blood and using isolated cells to measure LC3B-II protein. The method is valuable for evaluating intra-individual variation induced by interventions.

Summary: The tandem-tagged autophagy reporter mouse model is a useful tool for studying autophagy in the retina, but its application depends on various technical and biological factors, including microscope settings, transgene expression, choice of fluorophores, and the presence of lipofuscin autofluorescence.
