Summary: The study synthesized three new series of prenylated benzopyrans containing different numbers of isoprenoid units and explored their hPPAR transactivation activity and structure activity relationships. The results demonstrated that 2-prenylated benzopyrans with seven-carbon and nine-carbon side chains are good lead compounds for preventing cardiovascular diseases.

Thrombus formation during ECMO: Insights from a detailed histological analysis of thrombus composition

Summary: Intra-device thrombosis is a common complication during ECMO. This study examined the molecular and cellular features of ECMO thrombi and identified two main types: RBC-rich and fibrin-rich. The location of the thrombosis did not influence its composition, except for higher levels of H3Cit found in pump and oxygenator thrombi. The role of NETs in thrombus formation was found to be minor. These findings have implications for improving anticoagulation strategies in ECMO.

Hybrid closure of ventricular septal defect and implantation of systemic right ventricular assist device

Summary: We present a case of a 50-year-old female patient who was readmitted with refractory systemic right ventricular failure. The patient had a history of Mustard procedure during childhood for transposition of the great arteries, resulting in a significant residual ventricular septal defect. We describe the use of hybrid closure of ventricular septal defect prior to ventricular assist device implantation.

Modulation of inflammatory M1-macrophages phenotype by valvular interstitial cells

Summary: In patients with aortic valve stenosis, there is a significant increase in circulating monocytes and plasma transforming growth factor beta-1 levels, along with high macrophage infiltration in the thickened spongiosa of aortic valves, but decreased expression of matrix metalloproteinase-9. Valvular interstitial cells cultured from aortic valve stenosis exhibit a profibrotic phenotype and can influence the phenotype of inflammatory macrophages.

Summary: The transcription factor Basonuclin 2 (BNC2) is found to play a key role in driving myofibroblast activation in liver fibrosis. Its expression is induced in fibrotic livers and it integrates pro-fibrotic stimuli to induce matrisome gene expression. BNC2 deficiency leads to reduced collagen deposition in the liver. Furthermore, the study identifies a potential drug target, CC-885, as a BNC2 inhibitor.

Summary: This study suggests that metabolic dysfunction-associated fatty liver disease (MAFLD) and liver fibrosis severity have an impact on left atrial (LA) structure and function. MAFLD patients with a high risk of F3-F4 liver fibrosis showed abnormalities in LA low-voltage areas, peak left atrial longitudinal strain, and volume, as well as a higher rate of atrial fibrillation (AF) recurrence.

Fingolimod treatment modulates PPARγ and CD36 gene expression in women with multiple sclerosis

Summary: This study aimed to evaluate the effects of Fingolimod treatment on PPAR and CD36 gene expression. The results showed that Fingolimod treatment increased HDL-Cholesterol and Apolipoprotein E levels, as well as leukocyte PPAR gamma and CD36 gene expression. Additionally, the variations in PPAR gamma and CD36 were significantly correlated during therapy and in patients with stable disease.

Summary: COVID-19 convalescent plasma (CCP) contains neutralising antibodies that may be useful as passive immunotherapy. CCP showed increased levels of IL-6 and TNF-alpha and lower levels of IL-10 compared to control plasma. Auto-Antibodies against type I IFNs in CCP were not associated with reported adverse events. Further studies are needed to define the clinical relevance of these findings.

Bile acids contribute to the development of non-alcoholic steatohepatitis in mice

Summary: This study demonstrates that alterations in enterohepatic bile acids significantly contribute to the development of non-alcoholic steatohepatitis in relevant preclinical models. Experimental modulation of bile acid composition restored perturbed FXR and TGR5 signaling and prevented non-alcoholic steatohepatitis and associated metabolic disorders.
