Reciprocal activating interaction between 6-sulfo LacNAc+ dendritic cells and NK cells

Dendritic cells (DCs) display an extraordinary capacity to induce T-cell responses providing the opportunity of DC-based cancer vaccination strategies. Additional findings indicate that DCs may also play a crucial role for the activation of natural killer (NK) cells, which are important effectors in innate antitumor immunity. However, studies investigating the interaction between native human DCs and NK cells are limited. Recently, we defined 6-sulfo LacNAc (slan) DCs as a major subpopulation of myeloid human blood DCs, which represent principal producers of the proinflammatory cytokines tumor necrosis factor-alpha and interleukin (IL)-12. Functional data revealed that slanDCs efficiently induce neoantigen-specific CD4(+) T cells and activate tumor-reactive cytotoxic T cells. When evaluating the crosstalk between slanDCs and NK cells in this study, we found that lipopolysaccharide (LPS)-activated slanDCs efficiently enhance NK cell CD69 expression and interferon (IFN)-gamma secretion. NK cell-mediated tumor-directed cytotoxicity was significantly improved by slanDCs. INK cell activation induced by slanDCs was critically dependent on IL-12. When investigating the impact of NK cells on the immunostimulatory capacity of slanDCs, we observed that they promote DC maturation. In addition, NK cells strongly enhanced the secretion of immunomodulatory IL-12 and reduced the release of immunosuppressive IL-10 by slanDCs. IFN-gamma and cell-to-cell contact contributed to these effects. Furthermore, data revealed that DC-NK cell crosstalk improves slanDC-mediated differentiation of naive CD4(+) T lymphocytes into IFN-gamma-producing Th1 cells. In conclusion, we demonstrate a reciprocal activating interaction between slanDCs and NK cells, which may play a pivotal role in the regulation of antitumor immunity. (C) 2008 Wiley-Liss, Inc.

Expression of Potential Targets for Cell-Based Therapies on Melanoma Cells

Summary: High expression of gp100, TRP2, and GD2 proteins was observed in melanoma tumor samples, with TRP2 and ABCB5 expression correlated with lower tumor thickness in primary tumors. However, protein expression levels showed no association with survival in advanced melanoma patients. Abundant surface expression of GD2 and HER2 in melanoma cell lines suggests potential targets for CAR- or TCR-cell therapies against melanoma.

This study suggests that anti-PSCA IgG4-TM holds promising potential as a multifunctional radio-/immunotheranostic tool.

Summary: Molecular imaging allows non-invasive investigation of biological and biochemical processes, providing important information for understanding disease processes and planning treatment strategies. Combining nuclear and optical probes offers increased sensitivity and complementary information. This review article discusses dual-labelled systems for bimodal nuclear and optical imaging, with a focus on radiometal-labelled molecules and metal complexes combined with fluorescent dyes. The clinical potential, especially for fluorescence-guided surgery, is assessed.

Immunotargeting of Cancer Stem Cells

Summary: Tumor cells exhibit functional heterogeneity, and cancer stem cells (CSCs) are a population of tumor cells with self-renewal and differentiation properties. CSCs are found in most solid and hematological tumors and are characterized by various markers. These markers can be used to develop tumor-specific antibodies, immune cells, vaccines, and immune responses to tumor cells, including CSCs. This review discusses CSC-directed immunotherapies, their clinical development, safety and efficacy improvements, and future strategies for enhancing anti-CSC immunotherapy.

While most biological investigations are currently carried out in vitro, in vivo applications are becoming more important. Radiolabeled nanogels have potential for imaging and therapy.
