Structural requirements for the inhibition of calcium mobilization and mast cell activation by the pyrazole derivative BTP2

Mast cells play a critical role in the development of the allergic response. Upon activation by allergens and IgE via the high affinity receptor for IgE (Fc epsilon RI), these cells release histamine and other functional mediators that initiate and propagate immediate hypersensitivity reactions. Mast cells also secrete cytokines that can regulate immune activity. These processes are controlled, in whole or part, by increases in intracellular Ca2+ induced by the Fc epsilon RI. We show here that N-(4-(3,5-bis(trifluoromethyl)-1Hpyrazol-1-yl)phenyl)-4-methyl-1,2,3-thiadiazole-5-carboxamide (BTP2), a pyrazole derivative, inhibits activation-induced Ca2+ influx in the rat basophil cell line RBL-2H3 and in bone marrow-derived mast cells (BMMCs), without affecting global tyrosine phosphorylation of cellular proteins or phosphorylation of the mitogen-activated protein kinases Erk1/2, JNK and p38. BTP2 also inhibits activation-induced degranulation and secretion of interleukin (IL)-2, IL-3, IL-4, IL-6, IL-13, tumor necrosis factor (TNF)-alpha, and granulocyte macrophage-colony stimulating factor (GM-CSF) by BMMCs, which correlates with the inhibition of Nuclear Factor of Activated T cells (NFAT) translocation. In vivo, BTP2 inhibits antigen-induced histamine release. Structure-activity relationship analysis indicates that substitution at the C3 or C5 position of the pyrazole moiety on BTP2 (5-trifluoromethyl-3-methyl-pyrazole or 3-trifluoromethyl-5methyl-pyrazole, respectively) affected its activity, with the trifluoromethyl group at the C3 position being critical to its activity. We conclude that BTP2 and related compounds may be potent modulators of mast cell responses and potentially useful for the treatment of symptoms of allergic inflammation. (C) 2011 Elsevier Ltd. Summary: The study showed that carnosine can protect macrophages from cell death and apoptosis induced by Aβ 1-42 oligomers, decrease oxidative stress, enhance macrophage phagocytic activity, and rescue fractalkine receptor CX3CR1. These new findings support the therapeutic potential of carnosine in the context of AD pathology, highlighting its multimodal mechanism of action.

Fluorescent Detection of Peroxynitrite Produced by Myeloid-Derived Suppressor Cells in Cancer and Inhibition by Dasatinib

Summary: Myeloid-derived suppressor cells (MDSCs) are immature myeloid cells that expand dramatically in cancer patients, leading to immunosuppression. MDSCs mediate immunosuppression by producing peroxynitrite (PNT), which inactivates immune effector cells. A fluorescent sensor called PS3 allows direct detection of PNT, and using this method, it was found that cancer mice and melanoma patients have higher levels of PNT due to increased numbers of MDSCs. The kinase inhibitor dasatinib can block PNT production by inhibiting phagocytosis and reducing MDSC numbers.

Synthesis of a fluorinated pyronin that enables blue light to rapidly depolarize mitochondria

Summary: Fluorinated analogues of pyronin B, a fluorophore, were synthesized and shown to react reversibly with primary amines. These analogues undergo oxidation in the presence of proteins, yielding fluorescent pyronins, and this oxidation step can be enhanced by blue light irradiation. The fluorinated analogue 2,7-difluoropyronin B was found to localize in mitochondria in living HeLa cells and depolarize the mitochondria upon blue laser excitation. This photochemical control of mitochondrial biology is attributed to the enhanced cellular production of reactive oxygen species.

Summary: Mitochondria are the central metabolic hubs in cells, relying on proteins encoded by nuclear DNA and mitochondrial DNA (mtDNA) for their function. The maintenance and distribution of mtDNA are crucial for proper mitochondrial function and are associated with mitochondrial diseases. mtDNA is organized into nucleoids that dynamically move and interact with each other. The replication and distribution of mtDNA nucleoids are regulated by the complex interplay of mitochondrial dynamics, ER-mitochondria contact sites, and cytoskeletal networks.
