Region 4 of the RNA polymerase σ subunit counteracts pausing during initial transcription

Summary: The study reveals that the absence of the s4 promoter-binding domain renders the RNAP holoenzyme ineffective in de novo transcription initiation. The sigma 4 domain stabilizes short RNA:DNA hybrids, suppresses pausing, and enhances RNAP processivity. The antipausing activity of sigma 4 is modulated by its interaction with other factors, ultimately favoring productive transcription initiation.

Summary: Technological breakthroughs in electron microscopy have made it possible to study biological macromolecular complexes with novel challenges, such as sample preparation and heterogeneous macromolecular assemblies. In this study, a V-shaped DNA origami template was used to position proteins, demonstrating a new approach for characterizing mechanosensitive proteins and expanding the range of potential targets for single-particle EM investigation. Validation with RNAP protein showed compatibility with cryo-EM sample preparation.

Summary: NOSO-502, a preclinical antibiotic candidate, shows activity against Enterobacteriaceae pathogens but some colistin-resistant bacteria may develop resistance to it, possibly through mutations in the crrB gene. This study highlights the importance of CrrAB in controlling the resistance pathway and the role of the kexD gene in NOSO-502 susceptibility.

Summary: Researchers discovered that a stress-response factor called sigma(B) from Mycobacterium tuberculosis can induce the oligomerization of RNA polymerase (RNAP), resulting in the formation of a supramolecular complex composed of eight RNAP units. This finding reveals how sigma(B) can regulate gene expression by modulating the assembly and hibernation of RNAP holoenzyme.

Summary: This study evaluated multiple treatment failures to modern antiretroviral therapy in HIV-infected individuals and found that approximately 4% of them were difficult to treat. The difficult to treat group, compared to the non-difficult to treat group, was characterized by older age, higher prevalence of AIDS, lower CD4+ cell count, and higher risk of treatment failure.
