Summary: Six new structurally diverse abietane diterpenoids, Pinuskesiols A-F (1-6), were isolated from Pinus yunnanensis resins. Spectroscopic and computational methods were used to determine their structures, including absolute configurations. All the compounds contain a carbonyl functionality at C-4, with 1 and 2 behaving as a lactone. Compound 3 has an isopropyl group attached to C-13 via an O-atom. Moreover, the presence of a Delta 5(6) double bond and a ketone at C-7 makes compound 2 a large conjugated system. Biological evaluation showed that compounds 1, 2, and 4 could concentration-dependently inhibit iNOS and COX-2 expression in lipopolysaccharide-induced RAW 264.7 cells, with compound 2 being the most active in COX-2 inhibition.

Summary: Seven capnellenes derived from Capnella imbricata were discovered, including three previously unknown compounds. The structures of these compounds were determined through spectroscopic analysis and comparison with literature data. The compounds showed promising anti-inflammatory activity, with capnellenes 2 and 5 demonstrating significant reductions in iNOS levels and capnellenes 1, 5, and 7 exhibiting inhibitions against COX-2 protein expressions. Oxygen-bearing functionalities at C-8 and C-10 were found to play critical roles in inhibiting iNOS protein induction. Principal component analysis confirmed these capnellane-based sesquiterpenes as potential anti-inflammatory agents targeting iNOS-related targets.

Summary: In this study, two novel ester prodrugs of Oxyresveratrol (OXY), oxyresveratrol tetrapropionate (OXY-TPr) and oxyresveratrol tetrabutyrate (OXY-TBu), were synthesized and found to enhance the membrane permeation ability of OXY. Among them, OXY-TAc showed the most significant anti-inflammatory activity, reducing the secretion of various pro-inflammatory mediators and suppressing the activity of inflammation-related enzymes. The results suggest that OXY-TAc has improved pharmacological effects compared to its parent compound OXY.

Summary: The activation of the inflammasome plays a critical role in the pathogenesis of systemic lupus erythematosus (SLE), and mesenchymal stem cells (MSC) have been shown to alleviate SLE by suppressing inflammasome activation. This study found that the NLRP3 inflammasome was activated in macrophages from SLE patients and mice, and its activation correlated with disease activity. After MSC transplantation, the severity of SLE was reduced, and NLRP3 inflammasome activation was inhibited. These findings suggest that MSC suppress inflammasome activation and provide a potential therapeutic target for SLE.
