Sequence Diversity of the Bla g 4 Cockroach Allergen, Homologous to Lipocalins, from Blattella germanica

Background: The cockroach allergen Bla g 4, a putative lipocalin, is known to exhibit frequent sequence variations. However, the previously reported cDNA sequences are truncated at the N terminus. This study was undertaken to investigate the mechanisms by which these sequence variations are generated. Methods: Rapid amplification of cDNA ends PCR and RT-PCR were performed to obtain the full sequence of the Bla g 4 cDNA, and PCR was also used to clone the Bla g 4 genomic DNA. In addition, Bla g 4 protein variants were analyzed by two-dimensional gel electrophoresis. Results: Nine additional amino acid residues at the N terminus of Bla g 4 were identified, and 2 genes encoding Bla g 4, both of which consisted of 5 exons, were cloned. Examination of 34 clones of Bla g 4 cDNA obtained by RT-PCR revealed 14 variants. In particular, Bla g 4 sequences showed frequent clusters of variations in residues 38-45, 61-82 and 144-163. Differences in cDNA sequences may imply that RNA sequences are edited after transcription. More than 10 spots were identified between pH 5 and 7 upon two-dimensional gel electrophoresis, indicating that multiple variants of Bla g 4 are produced at the protein level. Conclusions: Genetic polymorphisms among individual cockroaches, the existence of multiple genes and sequence variations caused by RNA editing produce sequence diversity of Bla g 4, which may influence its allergenicity. The sequence information obtained in this study will be helpful for the standardization of the cockroach allergen and thereby aid in the development of diagnostics and immunotherapeutics. Summary: This study examines the evolutionary origin, diverse lifestyles, and quick adaptation of astigmatic mites in human household environments using six high-quality assembled and annotated genomes. The research reveals that astigmatic mites evolved from Prostigmata, notorious plant pests, and rapidly diversified from soil oribatid mites. The study also highlights the rapid expansion of gene families via tandem gene duplications in astigmatic mites, providing genetic resources for adaptation to changing household environments. Additionally, massive horizontal gene transfer events occurred during the evolution of astigmatic mites, enabling detoxification and digestive functions and offering potential drug targets for pest control.

Diagnosis of Balamuthia mandrillaris Encephalitis by Thymine-Adenine Cloning Using Universal Eukaryotic Primers

Oak Pollen Allergy in Korea

Summary: Oak pollen allergy is a common allergenic condition worldwide, causing pollinosis. While the molecular properties of some component allergens have been identified, research on others is still lacking. Neglecting to study some oak component allergens may be due to their high cross-reactivity with birch. However, utilizing these culprit allergen molecules could enhance diagnostic sensitivity and effectiveness of immunotherapy. Sensitization to oak and birch pollen often leads to pollen food allergy syndrome with fruits and vegetables as triggers. Acorns and wood dust from oak trees can also cause allergic diseases. This article summarizes the distribution, taxonomy, and molecular characteristics of allergenic oak trees, as well as cross-reactivity, clinical aspects for diagnosis and immunotherapy, with a focus on the situation in Korea.

Parasites and blood-meal hosts of the tsetse fly in Tanzania: a metagenomics study

Summary: House dust mite is a common cause of atopic dermatitis in humans and dogs. Detection of serum IgE to allergens is commonly used for diagnosis, but cross-reactivity and non-specific reactivity can lead to misdiagnosis. By comparing human and canine IgE reactivities to mite component allergens, different sensitization patterns were found between allergic humans and dogs.

Production of Dermatophagoides farinae Having Low Bacterial Content Using Ampicillin

Summary: This study examines the effect of antibiotic treatment on the bacterial concentration in house dust mites and its allergenic properties. It reveals that ampicillin treatment significantly reduces the bacterial content and lipopolysaccharide levels in the mites, but does not alter the concentration of major allergens. Furthermore, the extract from ampicillin-treated mites reduces allergic airway inflammation in human bronchial epithelial cells.
