Summary: Familial aggregation of HL has been shown to have a genetic component. In this study, whole genome sequencing was performed on individuals with and without HL from pedigrees with multiple affected relatives. A total of 44 HL-risk variants were identified, including both coding and noncoding variants. The study also identified several novel variants and observed segregation patterns in most pedigrees.

Summary: Malignant tumors pose a serious threat to human health and life, as well as hinder economic development. Human leukocyte antigen (HLA) is the most complex known polymorphic system, which has been shown to be associated with tumor occurrence and development. HLA molecules can regulate tumor cell proliferation and inhibit anti-tumor immunity. This review summarizes the structure and function of HLA molecules, the polymorphism and expression of HLA in tumor tissue, the roles of HLA in tumor cells and tumor immunity, and the potential clinical application of HLA in tumor immunotherapy, aiming to provide relevant information for the development of anti-tumor immunotherapies involving HLA in the clinic.

Summary: Hodgkin lymphoma is a hematological malignancy that arises from germinal-center B cells and has good overall prognosis. However, treating relapsed or resistant disease remains challenging, despite high survival rates from current treatment techniques. The development of late malignancies after successful cure poses a major concern, especially in pediatric patients. It is crucial to develop biomarkers to stratify patients and determine optimal treatment regimens to balance survival rates and late consequences. This article reviews the epidemiology, risk factors, staging, biomarkers, treatments, and late development of secondary malignancies in Hodgkin lymphoma patients.

Summary: Hodgkin lymphoma (HL) is a rare type of lymphoma, with two subtypes: classical Hodgkin's lymphoma (cHL) and nodular lymphocyte predominant Hodgkin lymphoma (NLPHL). cHL is highly curable with first-line chemotherapy, but some patients may relapse or fail to respond. High dose chemotherapy followed by autologous hematopoietic stem cell transplant (AHSCT) is the standard treatment modality for chemo sensitive cHL. Targeted immunotherapy, novel agents, and CAR T-cell therapy are being explored as potential treatment options.

Recent advances in gene expression profiling analysis have been applied to study the microenvironment component in cHL, and therapies targeting both neoplastic cells and the cellular components of the background have been described.
