Multi-armed cationic cyclodextrin:poly(ethylene glycol) polyrotaxanes as efficient gene silencing vectors

A family of branched polyrotaxanes (bPRTx(+)), threaded with multiple cationic alpha-cyclodextrins (alpha-CDs) onto a multi-armed poly(ethylene glycol) (PEG) core, were synthesized and studied as gene silencing vectors. These bPRTx(+) formed stable, positively charged complexes with diameters of 150-250 nm at N/P ratios as low as 2.5. The bPRTx(+) materials were shown to have gene-silencing efficiencies comparable to those of Lipofectamine 2000 (L2k) and bPEI, while displaying similar toxicity profiles. The unique structure of these polyrotaxanes allows them to effectively condense and complex siRNA into nanoparticles at much lower N/P ratios than L2k or bPEI. These findings suggest that bPRTx(+) may be useful materials for gene therapy applications.

Prospective memory impairment in chronic heart failure: a replication study

Structure-function relationships of cholesterol mobilization from the endo-lysosome compartment of NPC1-deficient human cells by ß- CD polyrotaxanes

Summary: This study explores a new approach for treating Niemann-Pick Type C (NPC) using polyrotaxanes containing 2-Hydroxypropyl-β-cyclodextrin (HP-β-CD). The study found that polyrotaxanes can remove cholesterol from NPC patient cells, and the endcapping and threading efficiency of polyrotaxanes are directly related to cholesterol mobilization. Additionally, improving the solubility of the compounds can significantly enhance cholesterol mobilization.

Use of clinical chemistry health outcomes and PFAS chain length to predict 28-day rodent oral toxicity

Summary: Current literature suggests that PFAS carbon chain length may affect its toxicity, and statistical modeling can be used to predict toxicity and improve the efficiency of PFAS regulation development. In this study, data analysis and predictive modeling were conducted, showing significant differences in mean values for 11 out of 15 health outcomes. Simple linear regressions were used for two health outcomes, yielding statistically significant results. Comparison between the results of an actual dataset and a theoretically generated dataset indicated no significant differences in the mean values of the two health outcomes. Therefore, predictive statistical modeling can be used to predict health outcomes for PFAS exposure.

Optimizing high-throughput viral vector characterization with density gradient equilibrium analytical ultracentrifugation

Summary: Accurate characterization of capsid species is crucial for viral vector-based gene therapies and vaccines. The current standard method, sedimentation velocity analytical ultracentrifugation (SV-AUC), is often size-limited and requires specialized software for analysis. The density gradient equilibrium AUC (DGE-AUC) provides a simplified and high-resolution method for separating biologics of different densities. It offers improvements in sensitivity and can be used with larger viral particles. This serotype-agnostic method enables intuitive interpretation and analysis without the need for specialized software.

On the feasibility of a component-based approach to predict aerodynamic noise from high-speed train bogies

Summary: At speeds above 300 km/h, aerodynamic noise from the bogie area in high-speed trains becomes significant. Component-based models are attractive for predicting aerodynamic noise due to their fast and cheap calculations. This study evaluates the feasibility of applying these models to predict the aerodynamic noise from bogies and adjusts the empirical constants using measurements in a wind tunnel.

Comprehensive model of a rotating flexible wheelset for high-frequency railway dynamics

A semi-analytical model of a discretely supported railway track
