Blockade of SHP2 enhances the anti-fibrotic efficacy of IFN-gamma in myofibroblasts.

THSG affects extracellular matrix deposition, fibronectin expression, oxidative stress, and autophagy. Therefore, THSG may be a potential drug for the treatment of pulmonary fibrosis.

Summary: In this study, the potential of rutin in attenuating ECM regulation and EMT caused by TGF-beta and its underlying mechanisms were investigated. Rutin was found to inhibit TGF-beta-induced ECM-related genes and EMT processes. Furthermore, rutin attenuated lung fibrosis in a mouse model. Molecular docking analyses suggest that rutin inhibits SMAD-mediated fibrotic signaling pathways. These findings highlight the potential of rutin as a therapeutic option for fibrotic and cancer-related diseases induced by TGF-beta.

Summary: In this study, a novel peptide DR4penA was designed based on the structure-activity relationship and rational design. DR4penA showed higher anti-PF activity, antioxidant activity, and a longer half-life compared to the original peptide DR8. It was found that DR4penA attenuated bleomycin- and paraquat-induced PF, and its anti-PF activity was equivalent to that of pirfenidone. Furthermore, DR4penA suppressed the TGF-beta/Smad pathway in TGF-beta 1-induced A549 cells and paraquat-induced rats. This study suggests that DR4penA is a potential candidate for PF therapy.

Summary: In this study, the anti-fibrotic effects of derrone on TGF-beta 1-stimulated MRC-5 lung fibroblast cells and bleomycin-induced lung fibrosis were investigated. It was found that high concentrations of derrone increased cytotoxicity in MRC-5 cells, but low concentrations did not induce significant cell death. Derrone reduced the expression of TGF-beta 1, fibronectin, elastin, and collagen1 alpha 1 in TGF-beta 1-stimulated MRC-5 cells, and also attenuated fibrotic histopathological changes in bleomycin-treated mice.

Summary: This study aimed to compare the relative efficacy and safety of infliximab and its biosimilars in patients with active rheumatoid arthritis (RA) who showed an inadequate response to methotrexate (MTX). A Bayesian network meta-analysis was conducted, combining direct and indirect evidence from randomized controlled trials (RCTs) to compare the efficacy and safety of infliximab biosimilars and the originator product in this patient population. The results showed that there were no significant differences in ACR20 response rates and serious adverse events (SAEs) between infliximab biosimilars and the originator.

Summary: This study compared the efficacy and safety of tacrolimus and mycophenolate mofetil (MMF) as induction therapy for lupus nephritis (LN). The results showed that there was no difference between tacrolimus and MMF in terms of complete or partial remission rates. However, tacrolimus had a lower incidence of herpes zoster infection and a higher risk of serum creatinine elevation compared to MMF. Overall, 3 mg/d tacrolimus was proven to be an effective and safe treatment for LN.

Summary: This study aimed to identify differentially expressed genes (DEGs) of systemic lupus erythematosus (SLE) using gene expression-based computational methodologies.
