Anti-fibrotic effect of thalidomide through inhibiting TGF-β-induced ERK1/2 pathways in bleomycin-induced lung fibrosis in mice

This study is designed to confirm the anti-fibrotic effect of thalidomide on bleomycin-induced lung fibrosis in a mouse model and to identify whether this anti-fibrotic effect is associated with inhibition of the transforming growth factor-beta (TGF-beta)-induced extracellular signal-regulated kinase1/2 (ERK1/2). C57BL/6 female mice were administered blomycin sulfate. In cultured human lung fibroblasts, expressions of type I collagen, fibronectin, and either TGF-beta or IL-6 were measured after thalidomide treatment by reverse transcription-polymerase chain reaction (RT-PCR). Expressions of ERK1/2, type I collagen, fibronectin, and TGF-beta 1 from lung tissues of blomycin-induced mice and from mouse lung fibroblasts were evaluated using RT-PCR and western blotting. Thalidomide administration significantly inhibits TGF-beta 1 mRNA expression in a dose-dependant manner following administration of IL-6 and IL-6R. In the analysis of BAL fluids, total BAL inflammatory cell counts, TGF-beta 1, and IL-6 levels in thalidomide-treated mice were significantly reduced when compared with bleomycin-treated mice (p < 0.01, p < 0.01, and p < 0.001, respectively). Thalidomide inhibited total ERK1/2 and phospho-ERK1/2 expression after TGF-beta 1 stimulation in the RT-PCR and western blotting. The results of our study suggest that the anti-fibrotic effect of thalidomide on lung fibrosis may be related to suppression of the TGF-beta 1-induced ERK1/2 signaling pathway.

Comparative efficacy and safety of infliximab and its biosimilars in patients with rheumatoid arthritis presenting an insufficient response to methotrexate A network meta-analysis

Summary: This study aimed to compare the relative efficacy and safety of infliximab and its biosimilars in patients with active rheumatoid arthritis (RA) who showed an inadequate response to methotrexate (MTX). A Bayesian network meta-analysis was conducted, combining direct and indirect evidence from randomized controlled trials (RCTs) to compare the efficacy and safety of infliximab biosimilars and the originator product in this patient population. The results showed that there were no significant differences in ACR20 response rates and serious adverse events (SAEs) between infliximab biosimilars and the originator.

A network meta-analysis of randomized controlled trials comparing the effectiveness and safety of voclosporin or tacrolimus plus mycophenolate mofetil as induction treatment for lupus nephritis

Summary: This study compared the effectiveness and safety of voclosporin + MMF, tacrolimus + MMF, and monotherapy for induction treatment in lupus nephritis patients. The results showed that tacrolimus + MMF and voclosporin + MMF were more effective than monotherapy, with tacrolimus + MMF being the most effective. However, tacrolimus + MMF had a higher risk of serious adverse events compared to monotherapy.

Relative efficacy and safety of Janus kinase inhibitors for the treatment of active psoriatic arthritis: a network meta-analysis

Summary: This study evaluated the effectiveness and safety of Janus kinase (JAK) inhibitors in patients with active psoriatic arthritis (PsA). The results showed that Filgotinib 200 mg and Upadacitinib 30 mg were the most effective in terms of ACR response rates, while Tofacitinib 10 mg was the most effective treatment based on PASI75. However, there were no significant differences in adverse events (AEs) and serious adverse events (SAEs) among the treatment options.

Comparative Efficacy and Safety of Biological Agents in the Treatment of Lupus Nephritis: A Network Meta-Analysis

Efficacy and safety of tacrolimus versus mycophenolate mofetil as induction treatment and low-dose tacrolimus as treatment for lupus nephritis: a meta-analysis

Summary: This study compared the efficacy and safety of tacrolimus and mycophenolate mofetil (MMF) as induction therapy for lupus nephritis (LN). The results showed that there was no difference between tacrolimus and MMF in terms of complete or partial remission rates. However, tacrolimus had a lower incidence of herpes zoster infection and a higher risk of serum creatinine elevation compared to MMF. Overall, 3 mg/d tacrolimus was proven to be an effective and safe treatment for LN.

Circulating leptin level in osteoarthritis and associations between leptin receptor polymorphisms and disease susceptibility: A meta-analysis

Summary: This study aimed to systematically review the relationship between circulating leptin levels and osteoarthritis (OA), as well as the association between leptin receptor (LEPR) polymorphisms and OA susceptibility. Meta-analysis results showed that patients with OA had significantly higher levels of circulating leptin and synovial leptin compared to healthy individuals. Additionally, the LEPR rs1137101 polymorphism was associated with an increased risk of developing OA.

Circulating leptin and its correlation with rheumatoid arthritis activity: a meta-analysis

Summary: This study investigated the association between leptin levels in the blood of individuals with rheumatoid arthritis (RA) and the severity of the disease. The results showed that RA patients had elevated leptin levels, which were correlated with the activity of RA. Characterization of virus-mediated autoimmunity and the consequences for pathological process in patients with systemic lupus erythematosus

Summary: This study aimed to identify differentially expressed genes (DEGs) of systemic lupus erythematosus (SLE) using gene expression-based computational methodologies. A total of 39 upregulated genes and 57 downregulated genes were identified in SLE patients compared to healthy controls. These DEGs were implicated in immune response pathways and were also associated with various virus infections. Additionally, certain genes were found to be closely associated with the quantities of immune cells.

Summary: This study aims to evaluate the standardized mortality ratios (SMRs) in patients with systemic sclerosis (SSc). The overall SMR of SSc patients is higher than that in the general population, and the SMR is significantly increased in both men and women. The mortality rate is also higher in diffuse cutaneous SSc (dcSSc) compared to limited cutaneous SSc (lcSSc).
