Summary: The study found that SubAB toxin induces cell apoptosis by significantly upregulating KLHDC7B expression. Knockdown of KLHDC7B inhibits the apoptotic pathway stimulated by SubAB, reducing cytotoxicity. Additionally, KLHDC7B regulates the expression of the HRK gene, affecting apoptosis caused by toxin-mediated ER stress.

Summary: The fluorescence imaging technique is widely used in natural and life sciences, but current commercial biomarkers have several disadvantages. In this study, two new fluorescent molecular rotors (SB-B2 and SB-B23) derived from Boron Schiff bases (BOSCHIBAs) were synthesized and characterized. The crystal structure of SB-B23 showed a binuclear complex. Both compounds exhibited viscochromic and thermochromic properties. These low cytotoxicity BOSCHIBAs showed selective fluorescent staining of the endoplasmic reticulum in HeLa cells and HUVECs.

Summary: The study found that in the absence of SubB, SubA is internalized into the endoplasmic reticulum (ER) and cleaves the chaperone GRP78, leading to cytotoxic effects. By analyzing a SubA mutant, the C-terminal SEEL motif was identified as an ER-targeting signal.

Summary: Mechanical strain induces the formation of ER exit sites and accelerates ER-to-Golgi trafficking. Impaired ER exit sites function results in reduced ability of cells to expand their surface area under mechanical stress and increases susceptibility to plasma membrane defects. Rac1 plays a crucial role in linking mechanical strain to ER export.

Summary: This study demonstrated that tunicamycin can induce the apoptosis of HSCs by increasing intracellular Ca2+ concentration and enhancing the expression of ERS proteins, while decreasing the expression of antiapoptotic proteins. The findings suggest that tunicamycin induces HSC apoptosis through the calpain-2/Ca2+-dependent ERS pathway.

Summary: Endometrial epithelial cells with mutations, including PIK3CA, showed a synergistic effect between insulin signaling and PIK3CA(H1047R) expression, resulting in differential gene expression and activation of interferon signaling and the unfolded protein response. Insulin treatment in mutated cells activated programs necessary to avoid endoplasmic reticulum stress-induced apoptosis. The overexpression of Viperin was identified as necessary but not sufficient for the synergistic effect of PIK3CA(H1047R) and insulin treatment.

Summary: This study reveals squalene epoxidase (SQLE) as a novel oncogene that promotes pancreatic cancer (PC) growth by mitigating endoplasmic reticulum stress and activating lipid raft-regulated Src/PI3K/Akt signaling pathway. SQLE facilitates cell proliferation, induces cell cycle progression, and inhibits apoptosis in vitro, while promoting tumor growth in vivo. SQLE inhibitors effectively suppress PC cell proliferation and xenograft tumor growth, highlighting the potential of SQLE as a therapeutic target for PC.

Summary: Prion diseases are a group of neurodegenerative disorders caused by the conversion of prion protein to its abnormal isoform. Studies have shown that ER stress and proteasome impairment occur early in spontaneous prion diseases, contributing to the accumulation of misfolded proteins. The findings suggest a strong correlation between ER stress, proteasome impairment and the neuropathological markers of prion diseases.

Summary: Deficiencies in mitochondrial import result in the toxic accumulation of non-imported precursor proteins, with some being redirected to the endoplasmic reticulum (ER). The ER delivery of endogenous mitochondrial trans-membrane proteins, particularly from the SLC25A family, is dependent on the GET complex. Loss of the GET pathway poses detrimental effects on yeast cells experiencing mitochondrial import failure.
