Summary: This study aimed to improve the accuracy of differential diagnosis of active tuberculosis (ATB) and latent tuberculosis infection (LTBI) by using fluorescent immunospot (FluoroSpot) assay to detect specific Th1 cell immune responses. Rv1733c SLP, combined with ESAT-6 and CFP-10, might be used as a candidate antigen for T cell-based tuberculosis diagnostic tests to differentiate ATB from LTBI.

Summary: The study identified cytokine biosignatures characterizing active and latent tuberculosis in children and found specific cytokines with significant expression differences between different groups of children. The potential of serum-based host biosignatures to aid in the diagnosis of childhood M.tb infections was highlighted, with proposed panels of markers showing promise in distinguishing between infected and uninfected children, as well as those with active TB and latent TB.

Summary: This study aimed to investigate the accuracy and utility of commercially available antibodies of Mycobacterium tuberculosis in the diagnostic assessment of suspected tuberculosis in high-burden countries. The results showed that commercially available antibodies have high specificity but low sensitivity for diagnosing active tuberculosis.

Summary: The study revealed that individuals with latent tuberculosis had significantly higher levels of IgA memory B cells specific to the antigen Rv2659c, and in the LTBI group, the number of IgA+ classical memory B cells was significantly higher than IgG+ memory B cells.

Summary: BCG vaccination induces similar immune responses to PPD but different responses to ESAT6/CFP10 in infants from Uganda and the UK. Maternal LTBI and infant exposure or infection with mycobacteria do not impact the immune response to BCG. Global differences in immune response to BCG are likely due to other factors.

Summary: This study analyzes peripheral blood samples from experimentally infected rhesus macaques, and identifies biomarkers that are associated with the activity and latency of tuberculosis. These biomarkers can predict bacterial load and differentiate between different states of tuberculosis infection.

Summary: Tuberculosis is a chronic infectious disease caused by Mycobacterium tuberculosis. While the BCG vaccine has been widely administered, its protective rate in preventing tuberculosis is not high in some regions, especially in adults. Therefore, research on the Mtb mechanism and development of new vaccines are crucial strategies. Recent studies have highlighted the role of PE and PPE proteins in the pathogenesis and chronic infection observed in TB, leading to the development of vaccines based on Mtb antigens.

Summary: This exploratory study aimed to identify biomarkers associated with active tuberculosis (TB). Through analyzing the plasma levels of inflammation-related proteins, a protein module strongly associated with active TB was identified. A 12-protein plasma signature was derived from the module and was found to be highly enriched in individuals with pulmonary and extrapulmonary TB, and associated with disease severity.

Summary: Inadequate tuberculosis diagnostics, especially in discriminating between active TB and latent TB infection, are major obstacles in disease burden reduction. The study investigated the role of lymphocyte non-specific function detection for TB diagnosis. Results show that CD4(+) T cell non-specific function has the potential for differentiating T-SPOT results, and the three-index diagnostic model is a potent tool for identifying different events in TB infection.
