Summary: This study compared the effects of different culture media on the expansion and characteristics of bone marrow-derived mesenchymal stromal cells (MSCs). The results showed that using xeno-free (XF) culture medium can more efficiently expand MSCs from MDS patients and improve their characteristics in in vitro and in vivo models.

Summary: The study evaluates the influence of ambient temperature, donor physiological status, and tissue source on sheep mesenchymal stem cells. Results show that adipose tissue-derived MSCs from full term pregnant female sheep exhibit higher potential for proliferation and differentiation compared to male sheep-derived MSCs. Cryopreserved MSCs have similar characteristics to fresh cells. Therefore, it is important to consider tissue type and donor physiological status when applying MSCs in clinical settings.

Summary: This study found that conditioned medium obtained from human tonsil-derived MSCs (T-MSC CM) can inhibit adipocyte differentiation by regulating glucocorticoid signal transduction, thereby reducing obesity. The experiments showed that T-MSC CM can attenuate early-stage adipocyte differentiation and protect mice from the negative effects induced by glucocorticoid treatment. Additionally, T-MSC CM enhances megakaryocyte differentiation and protects blood platelet counts.

Summary: The aim of this study was to provide an improved in vitro model of porcine monocyte-derived macrophages (MDMs) based on reduced-serum culture. The study assessed the impact of different culture conditions on the morphology, surface marker expression, and functional activities of MDMs, as well as the induction of cytokine gene expression. The results showed that reduced-serum culture conditions had a significant impact on the characteristics and function of MDMs. Furthermore, the study found that the choice of fetal bovine serum (FBS) supplier also influenced the phenotype and gene expression profile of MDMs.

Summary: Bone regeneration is a complex process involving the coordination of osteogenesis and osteoclastogenesis, with the balance between osteogenesis and adipogenesis of BMSCs playing a critical role. Recent studies have shown that exosomes from M2 macrophages facilitate osteogenesis and reduce adipogenesis through the miR-690/IRS-1/TAZ axis, potentially serving as a therapeutic tool for bone loss diseases.

Summary: In this study, it was found that hypoxic BMSCs-derived exosomes (hypBMSC-Exo) could alleviate cisplatin-induced ototoxicity and had better therapeutic effects compared to BMSC-Exo. This may be achieved by up-regulating the expression of SOD1 and SOD2 to protect auditory function.

Summary: Small extracellular vesicles (sEVs) play a critical role in cardiac cell therapy by delivering molecular cargo and mediating cellular signaling. Knocking down miR-192-5p and miR-432-5p in cardiac c-kit+ cell-derived sEVs enhances their therapeutic capabilities and improves cardiac function, reducing fibrosis and necrotic inflammatory responses. This study suggests that targeting deleterious miRNAs in sEVs could be a promising therapeutic strategy for chronic myocardial infarction.

Summary: Both adipose-derived and bone marrow-derived mesenchymal stem cells (ADSCs and BMSCs) can suppress renal fibrosis, with ADSCs showing a stronger effect under serum-containing culture conditions. However, ADSC infusion in mice may lead to a higher death rate from pulmonary embolism. Therefore, the selection of ADSCs or BMSCs for clinical applications should consider the target disease and culture method.

Summary: This study demonstrates that bone marrow-derived mesenchymal stem cells (MSCs) have vasculogenic properties. Different populations of MSCs vary in angiogenic factor expression and vasculogenic properties. The vasculogenic MSCs (vMSCs), distinct from endothelial progenitor cells (EPCs), may contribute to neovascularization and could be a promising cellular therapy for cardiovascular diseases.

Summary: ASS1 acts as a tumor suppressor in breast cancer, and SPA and LM-2I can suppress breast tumor cell proliferation and growth by activating ASS1. Treatment with SPA and LM-2I significantly enhances ASS1 enzymatic activity in breast cancer cells, especially those with low ASS1 expression, leading to reduced pyrimidine synthesis and inhibition of cancer cell proliferation.

Summary: As nano medications have advanced in the last four decades, nano-delivery systems have been used in the treatment of various diseases, particularly in cancer treatment. These systems can target cancer cells, improve treatment accuracy and effectiveness, and reduce systemic side effects. Among lipid-based nanoparticles, the load system shows great promise due to its high biocompatibility, biodegradability, loading capability, and immunogenicity. While previous reviews have focused on their effects and progress, this article highlights the importance of practical applications, such as intake methods, in determining the biological process of drugs. It also discusses potential solutions to problems that need further study in the future.
