Estrogens are well-known regulators of the immune responses. Most of their effects are mediated by two receptors: estrogen receptor (ER)alpha and ER beta. Up to date the presence of intracellular ER in human immune cells represents a controversial issue, while their surface membrane expression has scarcely been explored. In this study we investigated the intracellular and cell surface expression of ER alpha and ER beta in human peripheral blood lymphocytes (PBL) by flow and static cytometry as well as by Western Blot. To this aim we used five different commercial antibodies recognizing distinct ER epitopes. We observed that CD4(+) and CD8(+) T lymphocytes, B lymphocytes and NK cells contain intracellular ER alpha and ER beta, being the ER alpha 46 isoform the most represented ER. However, significant differences could be observed among the antibodies studied in terms of immunoreactivity and specificity. Importantly, we also found a cell surface expression of ER alpha 46 isoform. We also observed that a membrane-impermeant form of E2 induced a rapid phosphorylation of extracellular signal-regulated kinase (ERK), a significant proliferation of T lymphocytes, and IFN-gamma production by NK cells, thus suggesting the expression of a functional mER alpha. In conclusion our data could provide new insights as concerns the estrogen-related mechanisms of immune system modulation. They also suggest the need for a reappraisal of the experimental conditions for the characterization of the ER expression. (C) 2010 Elsevier B.V. All rights reserved.

Summary: Autophagy plays a crucial role in the activation of salivary gland epithelial cells (SGECs) in the pathogenesis of primary Sjogren's syndrome (SS). Autophagy activation is correlated with histologic severity and has been confirmed in both in vitro and in vivo experiments. Therefore, autophagy may serve as a new therapeutic target for primary SS.

A Soft Zwitterionic Hydrogel as Potential Coating on a Polyimide Surface to Reduce Foreign Body Reaction to Intraneural Electrodes

Manuele Gori, Sara Maria Giannitelli, Gianluca Vadala, Rocco Papalia, Loredana Zollo, Massimo Sanchez, Marcella Trombetta, Alberto Rainer, Giovanni Di Pino, Vincenzo Denaro

Summary: The organic zwitterionic-poly(sulfobetaine methacrylate) [poly(SBMA)]-hydrogel has been demonstrated as an effective coating to reduce foreign-body reaction caused by intraneural electrodes. This hydrogel exhibits low fouling characteristics and can be successfully coated on Polyimide surfaces through covalent bonds, showing potential for long-term release of anti-fibrotic drugs. Its properties make it a promising diffusion-based delivery system for slow and controlled anti-inflammatory and anti-fibrotic drug release in vivo.

Summary: This article provides a broad review of the role of hormone regulation in gender medicine and gender differences across key clinical areas. The study highlights the influence of sex differences in immune response, cardiovascular diseases, neurological disorders, cancer, and COVID-19. It also emphasizes the importance of gender implications in occupational health and health policy, calling for more personalized clinical medicine and public health approaches.

Summary: Gender medicine is a new medical approach that studies the differences between women and men in terms of prevention, diagnosis, and the outcome of diseases. Migraines, the most common neurological illness, have a higher prevalence in women than in men. They have a significant impact on the psychophysical health and productivity of workers.

Summary: This study proposes an innovative strategy for anti-SARS-CoV-2 immune response using extracellular vesicles (EVs). By fusing SARS-CoV-2 N with Nef(mut) or a deletion mutant of Nef(mut), the researchers successfully induced immune responses in human cells. The findings demonstrate the potential of engineered EVs to elicit a strong immune response in human cells.

Inhibition of cholesterol transport impairs Cav-1 trafficking and small extracellular vesicles secretion, promoting amphisome formation in melanoma cells

Summary: In this study, the role of cholesterol transport in the endosomal degradative-secretory system was explored using the U18666A inhibitor in a metastatic human melanoma cell line (WM266-4). The results showed that U18666A induced a shift of Cav-1 from the plasma membrane to the endolysosomal compartment, affecting the formation and release of small extracellular vesicles (sEVs). The inhibitor also altered the protein composition of sEVs and reduced their transfer capacity on target cells.
