Regulation of T lymphocyte apoptotic markers is associated to cell activation during the acute phase of dengue

Dengue fever, a public health problem in Brazil, may present severe clinical manifestations as result of an increased vascular permeability and coagulation disorders. T cell activation is a critical event for an effective immune response against infection, including the production of cytokines. We aim to reveal mechanisms that modulate the virus-cell interaction, with an emphasis on cell death. Apoptosis is involved in lymphocyte homeostasis, contributes to the clearance of virus-infected cells but also may play a role in the pathogenesis. Phosphatidylserine exposure on CD8T lymphocytes from dengue patients support early apoptotic processes and loss of genomic integrity, observed by DNA fragmentation in T lymphocytes and indicating late apoptosis. These T cells express activation and cytotoxic phenotypes as revealed by CD29 and CD107a upregulation. Higher frequencies of CD95 were detected in T lymphocytes mainly in those with the cytotoxic profile (CD107a(+)) and lower levels of anti-apoptotic molecule Bcl-2, suggesting that both CD4(+) and CD8(+) T cell subsets are more susceptible to apoptosis during acute dengue. The analysis of apoptosis-related protein expression profile showed that not only molecules with pro-but also those with anti-apoptotic functions are overexpressed, indicating that survival mechanisms could be possibly protecting cells against apoptosis caused by viral, immune, oxidative and/or genotoxic stresses. These observations led us to propose that in dengue patients there is an association between T cell susceptibility to apoptosis and the activation state. The mechanisms for understanding the immunopathogenesis during dengue infection are discussed.(C) 2013 Elsevier GmbH. All rights reserved.

Summary: Brazil faced a significant dengue virus epidemic in 2019, and through the use of Oxford Nanopore sequencing, researchers successfully monitored and generated 227 novel genome sequences of DENV1-2 from 85 municipalities. The study revealed complex transmission patterns of DENV1-2, with lineage co-circulation and replacement, and estimated effective reproduction numbers and patterns of seasonality for two lineages within the DENV2 BR-4 clade. The surveillance outputs and training initiatives presented here demonstrate the potential of real-time portable sequencing for genomic surveillance of emerging viruses.

Summary: Several studies have provided evidence of West Nile virus (WNV) circulation in Brazil, but the genomic diversity and transmission dynamics of the virus in the country are still largely unknown. Recent genomic monitoring activities in horses have revealed the presence of WNV in different regions of Brazil. This study retrospectively screened WNV samples from encephalitic horses and identified two independent introduction events of the virus in Brazil. These findings emphasize the importance of increasing the priority of WNV genomic monitoring in horses with encephalitis.

Summary: The COVID-19 pandemic in Brazil is assessed using phylogenetic and phylogeographic analysis. Multiple viral lineages were found to be circulating in Brazil with links to importations from Europe. Large local transmission clusters were detected within the country, and variants of concern and variants under monitoring emerged and spread both within and outside Brazil. Genomic surveillance in Brazil enabled real-time assessment of the spread of emerging SARS-CoV-2 variants.

Summary: Arbovirus infections in Brazil, such as chikungunya, dengue, yellow fever, and Zika, are major public health concerns. However, the limited availability of genomic data hinders our understanding of these outbreaks. This study combines epidemiological analysis and portable genome sequencing to retrospectively describe the CHIKV epidemic in Minas Gerais between 2017 and 2021. The results show multiple introductions of CHIKV into the state, likely from other regions of Brazil, and highlight the importance of genomic surveillance in infectious disease control.

Summary: In this study, a cosmopolitan genotype of dengue virus serotype 2 was identified in a 56-year-old male patient from the state of Goias in Brazil using nanopore sequencing and phylogenetic analysis. Summary: The COVID-19 epidemic in Brazil was initially driven by the Gamma variant, but was later replaced by the Delta variant. Unlike in other countries, the rapid spread of Delta in Brazil did not lead to a significant increase in cases and deaths, which may be attributed to the combination of early vaccination campaign and natural immunity from prior infection with the Gamma variant. The study highlights the importance of strengthening genomic monitoring on a national level to detect the emergence and spread of other variants of concern.
