Summary: This study assessed the expression of insulin-like peptides (ILPs) and ovary ecdysteroidogenic hormone (OEH) in Aedes aegypti during a gonadotrophic cycle and further functionally characterized these peptides to understand their roles in regulating egg formation. The results showed that all ILPs and OEH were expressed during the cycle, and there were differences in their biological activities and insulin- insulin growth factor signaling (IIS) phosphorylation levels.

Summary: The IGF family of ligands, receptors, and binding proteins play critical roles in normal human physiology and disease states. IGF-I regulates cellular growth, differentiation, and survival to control overall body growth, while IGF-II has important effects prenatally and in tissue-specific roles in adults.

Summary: SNAT2 is an important amino acid transporter involved in amino acid accumulation, cellular osmolarity, and cell growth. A potent inhibitor of SNAT2 has been identified through high-throughput screening, with selectivity against other transporters. Combined with a glucose transport inhibitor, this compound can halt the proliferative growth of cancer cells.

Summary: This study evaluates the contribution of protein nutrition and the GH/IGF-I system to body growth regulation. The results show that a reduction in serum EAA concentration hinders Igf1 transcription and decreases serum IGF-I levels. Restoring serum IGF-I levels through recombinant IGF-I infusion rescues body growth, indicating the importance of EAAs for animal growth.

Summary: Maternal malnutrition can lead to foetal growth restriction and increase the risk of diseases in children. This study investigates the changes in placental nutrient transporters during maternal hypoglycaemia in different stages of gestation in rats. The results suggest that the increase in placental nutrient transporters during maternal hypoglycaemia reflects an adaptive response to optimize foetal nutrient supply and development during limited glucose availability.

Summary: Phthalate exposure in women has been associated with reduced ovarian reserve and oocyte retrieval. In this study, we investigated the effects of di-n-butyl phthalate (DBP) exposure on the insulin-like growth factor 1 (IGF1) signaling in the ovary and found that DBP reduced Igf1 and Igf1r expression, as well as the number of ovarian follicles. These findings provide insight into how phthalates could influence the ovarian reserve in females.

Summary: Activation of T cell receptor results in metabolic reprogramming and upregulation of cell surface transporters for increased nutrient uptake, facilitating T cell exiting quiescence. The role of intracellular pathways of amino acid biosynthesis, specifically the activity of asparagine synthetase (ASNS), in CD8(+) T cell activation is crucial. ASNS expression is initially low in activated CD8(+) T cells, impairing cell viability and function, but later mTOR-dependent signals allow for ASNS upregulation, enabling cells to function independently of extracellular asparagine.

Summary: It was found that deficiency of Ppm1k leads to reduced metabolism of branched-chain amino acids (BCAAs) and protects against obesity-induced glucose intolerance. Accumulation of branched-chain keto acids (BCKAs) inhibits glucose production in liver cells. It was observed that BCKAs suppress the activity of liver mitochondrial pyruvate carrier (MPC) and pyruvate-supported respiration. Selective suppression of pyruvate-supported gluconeogenesis was seen in Ppm1k-deficient mice and could be restored through pharmacological activation of BCKA catabolism by BT2. Furthermore, liver MPC was found to be most sensitive to circulating BCKA levels, making it a sensor of BCAA metabolism.

Summary: This study identified SLFN5 as a novel regulator of the LAT1 amino acid transporter in castration-resistant prostate cancer (CRPC), contributing to mTORC1 activity. High expression of SLFN5 in CRPC tumors was correlated with poor patient outcome, indicating its clinical relevance as a potential target for CRPC treatment.

Summary: Reduced STAT3 activity in mice leads to increased susceptibility to DSS-induced colitis, indicating that the IL-6/gp130/STAT3 signaling pathway orchestrates intestinal barrier function by modulating cytokine secretion and promoting epithelial integrity to maintain defense against bacteria. Therapeutically targeting this signaling axis may serve as a treatment strategy for IBD patients.

Summary: IL11 is a member of the IL6 family of cytokines, and plays a role in regulating CD4+ T cell-mediated antitumor responses by suppressing the activity of host CD4(+) T cells in the tumor microenvironment. This suggests a potential immunomodulatory role for IL11 in colon cancer development that could be targeted for anticytokine-based therapy.
