Gender-dependent differences in serum profiles of insulin and leptin in caloric restricted rats

In the present study, we have investigated whether differences between male and female rats described in response to 40% caloric restriction (CR) were influenced by circulating level variations of sex hormones and/or insulin and leptin. Body weights (BW), organ weights, and adipose depot weights (ADW) were also measured. The most affected tissues by CR were the fat depots. Metabolically active organs were the least affected, especially more in females than in males (male weight lost: 24.3%vs.female: 17.3%). Testosterone and estradiol circulating levels did not show changes by CR. Insulin levels were decreased by CR in both genders, but was more evident in female rats than males. Leptin serum levels were higher in male rats than in females, and CR caused a circulating leptin level reduction only in males. In conclusion, our results indicate that leptin and insulin could be one of the keys of the different hormonal control of energy homeostasis in response to CR between female and male rats. In this sense, leptin serum levels correlated statistically with BW and with individual ADW only in male rats, whereas insulin serum levels correlated statistically with BW and with any of the ADW studied only in females.

Summary: Metastasis is a crucial factor in the mortality of colorectal cancer (CRC) patients. This study explores the changes in mitochondrial function associated with the metastatic stage of CRC and investigates whether they could be utilized as therapeutic targets. The findings indicate notable differences in mitochondrial functionality between tumor tissue and normal adjacent tissue in metastatic CRC patients. These alterations contribute to the promotion and maintenance of the metastatic phenotype, highlighting the impairment of mitochondrial function in tumor tissue during the metastatic stage.

Inflammation- and Metastasis-Related Proteins Expression Changes in Early Stages in Tumor and Non-Tumor Adjacent Tissues of Colorectal Cancer Samples

Summary: Non-tumor adjacent tissue has an important role in the development and inflammatory process of colorectal cancer. This study analyzed the expression of inflammatory, epithelial-mesenchymal transition (EMT), and metastasis-related proteins in tumor and non-tumor adjacent tissues from colorectal cancer patients. The results showed that tumor tissue had increased levels of metastasis and EMT-related proteins compared to non-tumor adjacent tissue, particularly in stage II. Tumor tissue from stage II also had increased levels of inflammatory-related proteins, which was also observed in non-tumor adjacent tissue from stage II. Additionally, higher expression levels of Vimentin and VEGF-B were associated with lower relapse-free survival in stage II patients. These findings suggest that these proteins could be potential biomarkers for early diagnosis of colorectal cancer.

Inflammation-Related Signature Profile Expression as a Poor Prognosis Marker after Oxaliplatin Treatment in Colorectal Cancer

Summary: This study analyzed the effect of oxaliplatin on tumor spheres formation efficiency, cell viability, cancer stem cells, and inflammation in primary- and metastatic-derived colorectal tumor spheres. The results showed that primary-derived spheres responded to oxaliplatin by modulating cancer stem cells and stemness properties, while metastatic-derived spheres exhibited an inflammatory response. In addition, the expression of inflammatory markers after oxaliplatin treatment correlated with poor prognosis and a metastatic phenotype. These findings highlight the importance of drug testing and personalized medicine in early-stage colorectal cancer.

Isolation and Characterization of Extracellular Vesicles in Human Bowel Lavage Fluid

Summary: Colorectal cancer is the third most common cancer worldwide, and the lack of early and specific biomarkers leads to late-stage detection. Tumors release extracellular vesicles (EVs) that have various functions and can be used as a source of biomarkers. Bowel lavage fluid (BLF) obtained during colonoscopy is an underutilized sample that represents EVs from tumor cells and has the potential to be a useful research tool. In this study, EVs from BLF were isolated and characterized, confirming their suitability for CRC study and potential as biomarkers for diagnosis and monitoring.

Growing underground: Development of thermogenesis in pups of the fossorial rodent Ctenomys talarum

Summary: In mammals, the integrated requirements of all activities must conform to a sustained rate of metabolism for the pup's development and adult life. Partitioning available energy according to short-term priorities allows animals to optimize long-term reproductive success. In this study, the development of thermogenic capacity in altricial rodent pups, specifically Ctenomys talarum, was evaluated. The results showed that non-shivering thermogenesis was absent in the pups until day 60, despite the presence of molecular markers for thermogenesis in brown adipose tissue deposits. However, the total thermogenic capacity became fixed at an early age, indicating an improvement in shivering thermogenesis efficiency after day 10.
