Histone deacetylase 1, 2, 6 and acetylated histone H4 in B- and T-cell lymphomas

Histone deacetylase (HDAC) inhibitors are novel therapeutics in the treatment of peripheral T-cell lymphoma, unspecified (PTCL) and diffuse large B-cell lymphoma (DLBCL), where, for unknown reasons, T-cell malignancies appear to be more sensitive than B-cell malignancies. The aim was to determine HDAC expression in DLBCL and PTCL which has not previously been investigated. The expression of HDAC1, HDAC2, HDAC6 and acetylated histone H4 was examined immunohistochemically in 31 DLBCL and 45 PTCL. All four markers showed high expression in both DLBCL and PTCL compared with normal lymphoid tissue. HDAC1 was more abundantly expressed in PTCL than in DLBCL (P = 0.0046), whereas acetylated H4 was more frequent in DLBCL (P < 0.0001), the latter suggesting a mechanism for T-cell lymphoma sensitivity to HDAC inhibitors. Moderate to strong HDAC6 expression was significantly correlated with favourable outcome (P = 0.016) in DLBCL patients, whereas the opposite effect was observed in PTCL patients (P < 0.0001). The other markers did not correlate with survival (P > 0.05). HDAC1, HDAC2, HDAC6 and acetylated H4 are overexpressed in DLBCL and PTCL relative to normal lymphoid tissue. Furthermore, HDAC6 may be an important prognostic marker associated with favourable outcome in DLBCL and a more aggressive course in PTCL.

However, the vaccines did not lead to clinical responses, but the study indicates that vaccines targeting mutant CALR may have potential in enhancing anti-tumor immune responses.

Summary: This prospective study found a sensitivity of 66% for diagnosing lymphoma with a core needle biopsy. The core needle biopsy method was correct in all non-lymphoma cases, retaining a specificity of 100%. However, in 18% of cases, the core needle biopsy method gave an incorrect lymphoma subtype.

Summary: Cutaneous T-cell lymphoma (CTCL) is a devastating lymphoid malignancy characterized by the accumulation of malignant T cells in the dermis and epidermis. Skin lesions cause serious symptoms and increase the risk of bacterial infection. The study found that malignant T cells secrete cytokines that downregulate the expression of filaggrin and filaggrin-2, leading to compromised skin barrier. Treatment with JAK inhibitors can reverse this effect and increase filaggrin expression in patients with advanced CTCL.

Summary: In this study, researchers found that FASN is highly expressed in CTCL cells, and inhibition of FASN affects cell viability and proliferation. The upregulation of FASN induced by FASN inhibitors can be partially reversed by SREBP. Therefore, the combination of FASN and SREBP inhibitors may have promising therapeutic effects in CTCL.

Summary: Using NanoString technology, we measured gene expression during wound healing and identified 22 upregulated and 6 downregulated differentially expressed genes. These findings correlate well with the current understanding of physiological reepithelialization and may provide potential therapeutic targets for patients with impaired healing.

Lymphomas of the salivary glands: a systematic review

Depending on the anatomical location, the lymphoma subtypes vary in aggressiveness, stage, and prognosis.
