Inflammation and Insulin Resistance as Risk Factors and Potential Therapeutic Targets for Alzheimer's Disease

Angeles Vinuesa, Carlos Pomilio, Amal Gregosa, Melisa Bentivegna, Jessica Presa, Melina Bellotto, Flavia Saravia, Juan Beauquis

Summary: Overnutrition and diets high in saturated fat contribute to inflammation and metabolic disturbances, leading to an increased risk for conditions like Alzheimer's disease. Understanding the interplay between inflammation and insulin resistance may offer potential therapeutic targets to prevent neurodegeneration and cognitive impairment.

A metabotropic glutamate receptor 3 (mGlu3R) isoform playing neurodegenerative roles in astrocytes is prematurely up-regulated in an Alzheimer's model

Juan Turati, Julieta Rudi, Juan Beauquis, Lila Carniglia, Federico Lopez Couselo, Julieta Saba, Carla Caruso, Flavia Saravia, Mercedes Lasaga, Daniela Durand

Summary: The altered expression of mGlu3 Delta 4 in Alzheimer's disease (AD) is associated with the onset of the disease, and it has a detrimental effect on astrocytes. The expression levels of mGlu3 Delta 4 increase with age, and they are prematurely overexpressed in the hippocampus of PDAPP-J20 mice. Moreover, heterologous expression of mGlu3 Delta 4 leads to cell death in astrocytes.

Diabetic patients treated with metformin during early stages of Alzheimer's disease show a better integral performance: data from ADNI study

Carlos Pomilio, Nicolas Gonzalez Perez, Ismael Calandri, Lucia Crivelli, Ricardo Allegri, Gustavo Sevlever, Flavia Saravia

Summary: We evaluated the effect of the antidiabetic drug metformin on patients with mild cognitive impairment due to Alzheimer's disease. The study found that metformin treatment improved cognitive performance, CSF biomarkers profile, and neuroanatomical measures in these patients.
