Summary: Aberrant amino acid metabolism, particularly the excessive plasma kynurenine (Kyn), is common in obesity. This study demonstrates that overexpressed indoleamine 2,3-dioxygenase 1 (IDO1) in adipocytes is the primary source of excessive Kyn, highlighting the central role of adipocytes in Kyn metabolism. Depletion of Ido1 in adipocytes eliminates Kyn accumulation, protecting mice against obesity.

Summary: Previous studies have shown that the interaction between brain-metastasizing melanoma cells and microglia plays a crucial role in promoting metastasis. In this study, we conducted a comprehensive investigation of melanoma-microglia interactions and discovered a pro-metastatic molecular mechanism that drives the melanoma-brain metastasis cycle. By analyzing different human brain-metastasizing melanoma cell lines, we found that exposure of microglia cells to melanoma-derived IL-6 resulted in increased STAT3 phosphorylation and SOCS3 expression, leading to enhanced melanoma cell viability and metastatic potential. Inhibitors targeting the IL-6/STAT3 pathway diminished the pro-metastatic functions of microglia and reduced melanoma progression. Furthermore, overexpression of SOCS3 in microglia cells increased melanoma cell migration and proliferation, providing support for melanoma brain metastasis. However, different melanomas exhibited heterogeneity in their ability to activate microglia and respond to microglia-derived signals. Overall, the activation of the IL-6/STAT3/SOCS3 pathway in microglia appears to be a major mechanism by which melanoma-microglia signaling contributes to the progression of melanoma brain metastasis.

Summary: This study reveals that early-stage infection with various viruses, including influenza A virus, leads to IL-6-independent phosphorylation of STAT3 Y705, which is critical for antiviral immunity. Knockin mice with STAT3Y705F/1 mutation show suppressed antiviral response, lung tissue injury, and poor survival upon IAV infection. The phosphorylation of STAT3 Y705 restrains IAV pathogenesis by repressing excessive IFN production, and blocking the phosphorylation enhances IFN expression and potentiates IAV virulence in mice. Knockout of IFNAR1 or IFNLR1 in STAT3Y705F/1 mice protects them from lung injury and reduces viral load.

BHE downregulates the expression of conventional dendritic cells-2 (cDC2) and cyclin B1 while upregulating the expression of myelin transcription factor 1 (MyT1). These findings provide a theoretical basis for the potential therapeutic use of blue honeysuckle extract as an inhibitor of HCC.

Summary: ICPC-a is a 45 kDa molecular weight extract from Imperata cylindrica, composed of α-D-1,3-Glcp and α-D-1,6-Glcp. It has thermal stability up to 220°C and exhibits an amorphous nature confirmed by X-ray diffraction analysis, with a layered morphology observed by scanning electron microscopy. ICPC-a significantly reduces uric acid levels, ameliorates uric acid-induced cell injury and apoptosis, and protects against renal injury by modulating multiple pathways including lipid peroxidation, antioxidant damage and defense levels, inflammatory factors secretion, purine metabolism, PI3K-Akt signaling pathway, NF-κB signaling pathway, inflammatory bowel disease, mTOR signaling pathway, and MAPK signaling pathway. These findings highlight ICPC-a as a promising natural substance for further research and development due to its multiple targets, pathways of action, and non-toxicity.

Preclinical evidence suggests that IL-6 antibodies may enhance the efficacy of immune checkpoint inhibitors (ICIs) by modulating the tumor microenvironment. However, more research is needed to determine the potential of IL-6 antibodies in overcoming resistance to ICIs in BTC.
