Gankyrin Promotes Tumor Growth and Metastasis Through Activation of IL-6/STAT3 Signaling in Human Cholangiocarcinoma

Although gankyrin is involved in the tumorigenicity and metastasis of some malignancies, the role of gankyrin in cholangiocarcinoma (CCA) is unclear. In this study we investigated the expression of gankyrin in human CCA tissues and cell lines. The effects of gankyrin on CCA tumor growth and metastasis were determined both in vivo and in vitro. The results showed that gankyrin was overexpressed in CCA tissues and cell lines. Gankyrin expression was associated with CCA histological differentiation, TNM stage, and metastasis. The multivariate Cox analysis revealed that gankyrin was an independent prognostic indicator for overall survival. Gankyrin overexpression promoted CCA cell proliferation, migration, and invasion, while gankyrin knockdown inhibited CCA tumor growth, metastasis, and induced Rb-dependent senescence and G(1) phase cell cycle arrest. Gankyrin increased the phosphorylation of signal transducer and activator of transcription 3 (STAT3) and promoted the nuclear translocation of p-STAT3. Suppression of STAT3 signaling by small interfering RNA (siRNA) or STAT3 inhibitor interfered with gankyrin-mediated carcinogenesis and metastasis, while interleukin (IL)-6, a known upstream activator of STAT3, could restore the proliferation and migration of gankyrin-silenced CCA cells. The IL-6 level was decreased by gankyrin knockdown, while increased by gankyrin overexpression. Gankyrin regulated IL-6 expression by way of facilitating the phosphorylation of Rb; meanwhile, rIL-6 treatment increased the expression of gankyrin, suggesting that IL-6 was regulated by a positive feedback loop involving gankyrin in CCA. In the xenograft experiments, gankyrin overexpression accelerated tumor formation and increased tumor weight, whereas gankyrin knockdown showed the opposite effects. The in vivo spontaneous metastasis assay revealed that gankyrin promoted CCA metastasis through IL-6/STAT3 signaling pathway. Conclusion: Gankyrin is crucial for CCA carcinogenesis and metastasis by activating IL-6/STAT3 signaling pathway through down-regulating Rb protein. (Hepatology 2014;59:935-946)

The Effective Analysis for Blue Honeysuckle Extract in the Treatment of Hepatocellular Carcinoma

Summary: This study demonstrated that blue honeysuckle extract (BHE) inhibits proliferation of hepatocellular carcinoma (HCC) cells by arresting the cell cycle in the G2/M phase. BHE downregulates the expression of conventional dendritic cells-2 (cDC2) and cyclin B1 while upregulating the expression of myelin transcription factor 1 (MyT1). These findings provide a theoretical basis for the potential therapeutic use of blue honeysuckle extract as an inhibitor of HCC.

Protocol for the identification and expression analysis of a cytoplasmic membrane-localized protein STING

Summary: In this article, a protocol for detecting two STING isoforms in human peripheral blood mononuclear cells or mouse splenocytes using Western blot and PCR is presented. The steps for constructing plasmids encoding each isoform and transferring them into mouse and human cell lines are described. Finally, the detection of cell membrane localization of pmSTING using flow cytometry, immunoprecipitation, and immunofluorescence is explained. Summary: ICPC-a is a 45 kDa molecular weight extract from Imperata cylindrica, composed of α-D-1,3-Glcp and α-D-1,6-Glcp. It has thermal stability up to 220°C and exhibits an amorphous nature confirmed by X-ray diffraction analysis, with a layered morphology observed by scanning electron microscopy. ICPC-a significantly reduces uric acid levels, ameliorates uric acid-induced cell injury and apoptosis, and protects against renal injury by modulating multiple pathways including lipid peroxidation, antioxidant damage and defense levels, inflammatory factors secretion, purine metabolism, PI3K-Akt signaling pathway, NF-κB signaling pathway, inflammatory bowel disease, mTOR signaling pathway, and MAPK signaling pathway. These findings highlight ICPC-a as a promising natural substance for further research and development due to its multiple targets, pathways of action, and non-toxicity.

The present roles and future perspectives of Interleukin-6 in biliary tract cancer

Summary: Biliary tract cancer (BTC) is a highly malignant tumor originating from the bile duct epithelium. Interleukin-6 (IL-6) plays a pivotal role in the development and progression of BTC, serving as a clinical biomarker for diagnosis and prognosis. Preclinical evidence suggests that IL-6 antibodies may enhance the efficacy of immune checkpoint inhibitors (ICIs) by modulating the tumor microenvironment. However, more research is needed to determine the potential of IL-6 antibodies in overcoming resistance to ICIs in BTC.
