Microcirculatory changes in the liver of patients with refractory ascites and their relationship with diabetes and alcohol

Laure Elkrief, Sophie Buyse, Xaviere Panhard, Clotilde Baudry, Nathalie Gault, Richard Moreau, Pierre-Emmanuel Rautou, Jacques Belghiti, Francois Durand, Pierre Bedossa, Valerie Paradis, Dominique Valla

Summary: The study found that changes in liver microcirculation are associated with refractory ascites, and diabetes and alcohol may explain refractory ascites by causing microangiopathy.

Characterization and Pharmacological Validation of a Preclinical Model of NASH in Gottingen Minipigs

Valerie Duvivier, Stepanie Creusot, Olivier Broux, Aurelie Helbert, Ludovic Lessage, Kevin Moreau, Nicolas Lesueur, Lindsay Gerard, Karine Lemaitre, Nicolas Provost, Edwige-Ludiwyne Hubert, Tania Baltauss, Angelique Brzustowski, Nathalie De Preville, Julia Geronimi, Lucie Adoux, Franck Letourneur, Adel Hammoutene, Dominique Valla, Valerie Paradis, Philippe Delerive

Summary: This study reports the development of a dietary model of NASH in the Gottingen minipig, which recapitulates most features of human NASH and may have improved translational value. The model was characterized using biochemical, histological, and transcriptional analyses, and the pharmacological response to treatment with Obeticholic acid was evaluated.

Paroxysmal nocturnal hemoglobinuria and vascular liver disease: Eculizumab therapy decreases mortality and thrombotic complications

Aurelie Plessier, Marina Esposito-Farese, Anna Baiges, Akash Shukla, Juan Carlos Garcia Pagan, Emmanuelle De Raucourt, Isabelle Ollivier-Hourmand, Jean-Paul Cervoni, Victor De Ledinghen, Zoubida Tazi, Jean-Baptiste Nousbaum, Rene Bun, Christophe Bureau, Christine Silvain, Olivier Tournilhac, Mathieu Gerfaud-Valentin, Francois Durand, Odile Goria, Luis Tellez, Agustin Albillos, Stefania Gioia, Oliviero Riggio, Andrea De Gottardi, Audrey Payance, Pierre-Emmanuel Rautou, Louis Terriou, Aude Charbonnier, Laure Elkrief, Regis Peffault de la Tour, Dominique-Charles Valla, Nathalie Gault, Flore Sicre de Fontbrune

Summary: The study aimed to assess the impact of eculizumab on the outcome of patients with paroxysmal nocturnal hemoglobinuria (PNH) and vascular liver disease (VLD). The results showed that eculizumab was significantly associated with lower mortality and less thrombotic recurrence in these patients, while other secondary outcomes did not reach statistical significance.

Contrast-enhanced CT and liver surface nodularity for the diagnosis of porto-sinusoidal vascular disorder: A case-control study

Shantha Ram Valainathan, Riccardo Sartoris, Laure Elkrief, Marta Magaz, Fabian Betancourt, Silvia Pellegrino, Arianna Nivolli, Marco Dioguardi Burgio, Yves Flattet, Sylvain Terraz, Nicolas Drilhon, Marie Lazareth, Julia Herrou, Onorina Bruno, Audrey Payance, Aurelie Plessier, Francois Durand, Maxime Ronot, Dominique-Charles Valla, Valerie Paradis, Juan Carlos Garcia-Pagan, Valerie Vilgrain, Pierre-Emmanuel Rautou

Summary: This study demonstrates that liver surface nodularity (LSN) and CT features are highly correlated with the diagnosis of Porto-sinusoidal vascular disorder (PSVD). A combination of LSN < 2.5 and normal-sized or enlarged segment IV strongly suggests PSVD in patients with signs of portal hypertension.

Clinical Interest of Serum Alpha-2 Macroglobulin, Apolipoprotein A1, and Haptoglobin in Patients with Non-Alcoholic Fatty Liver Disease, with and without Type 2 Diabetes, before or during COVID-19

Olivier Deckmyn, Thierry Poynard, Pierre Bedossa, Valerie Paradis, Valentina Peta, Raluca Pais, Vlad Ratziu, Dominique Thabut, Angelique Brzustowski, Jean-Francois Gautier, Patrice Cacoub, Dominique Valla

Summary: In patients with non-alcoholic fatty liver disease (NAFLD), type 2 diabetes mellitus (T2DM) may exacerbate the associations between alpha-2 macroglobulin (A2M), apolipoprotein Al (ApoA1), haptoglobin, liver fibrosis, inflammation, and COVID-19.

Usual transaminase values: should they be reviewed and adjusted?

Alexandre Raynor, Anna Soavelomandroso, Tiphaine Robert-Mercier, Dominique Valla, Katell Peoc'h, Celia Raulet-Bussian

Summary: The reference values for ALT and AST vary depending on the analytical method and individual characteristics, and it is important for clinical laboratories to update their reference values according to factors such as sex, body mass index, and age.

A new NRF2 activator for the treatment of human metabolic dysfunction-associated fatty liver disease

Adel Hammoutene, Samira Laouirem, Miguel Albuquerque, Nathalie Colnot, Angelique Brzustowski, Dominique Valla, Nicolas Provost, Philippe Delerive, Valerie Paradis

Summary: This study investigates the therapeutic potential of a new NRF2 activator, S217879, in treating human MAFLD and its underlying mechanisms. The results demonstrate that S217879 can improve lipid metabolism, activate antioxidant response, enhance autophagy, and reduce DNA damage, apoptosis, inflammation, and fibrogenesis.

MRI Phase-Contrast Blood Flow in Fasting Pediatric Patients with Fontan Circulation Correlates with Exercise Capacity

Christopher Z. Lam, Dawn David, Laura Acosta Izquierdo, Parneyan Pezeshkpour, Anne Dipchand, Emilie Jean-St-Michel, Rajiv R. Chaturvedi, Simon C. Ling, Rachel M. Wald, Govind B. Chavhan, Michael Seed, Shi-Joon Yoo

Summary: This study assessed regional blood flow in fasting pediatric patients with Fontan circulation using MRI and found that these patients had lower blood flow compared to healthy volunteers. Fasting Fontan blood flow correlated with pre-Fontan mean pulmonary artery pressure, ventricular end diastolic pressure, and post-Fontan percent predicted oxygen consumption at peak workload.

Hepatic venous pressure gradient in sinusoidal obstruction syndrome: diagnostic value and link with histological lesions

Simon B. Gressens, Dominique Cazals-Hatem, Virginie Lloyd, Aurelie Plessier, Audrey Payance, Didier Lebrec, Francois Durand, Gerard Socie, Dominique Valla, Valerie Paradis, David Michonneau, Pierre -Emmanuel Rautou

Summary: Liver sinusoidal obstruction syndrome (SOS) can be diagnosed using hepatic venous pressure gradient (HVPG) measurement, but low HVPG values do not rule out the presence of SOS, so HVPG should be combined with transjugular liver biopsy for the diagnosis of SOS.
