The molecular clock and central clock regulate the circadian rhythm, influencing lipid metabolism and vascular inflammation. This review summarizes experimental studies on the relationship between circadian rhythm, lipid metabolism, vascular inflammation, and atherosclerosis, as well as the effects of medication timing, bright light, and melatonin on cardiovascular health. The review also suggests future research agendas and provides recommendations for clinical practice.

The gut-brain vascular axis in neuroinflammation

Summary: The diverse microbiota in our gut plays a critical role in maintaining the immune, metabolic, and tissue homeostasis of the intestine and other organs, including the central nervous system. Microbial dysbiosis, characterized by a disrupted gut epithelial and vascular barrier function known as leaky gut, is associated with inflammatory intestinal diseases and may contribute to metabolic, inflammatory, and neurodegenerative diseases. Recent research has highlighted the strong connection between the gut and the brain through a novel vascular axis. This article aims to explore the gut-brain axis, focusing on the relationship between microbial dysbiosis, leaky gut, cerebral and gut vascular barriers, and neurodegenerative diseases such as Alzheimer's, Parkinson's, major depressive, and anxiety disorders. Understanding how disease pathophysiology, mucosal barrier function, and host-microbe interactions are related will enable the use of the microbiome as a biomarker for health and disease, as well as a target for therapeutic and nutritional advancements.

A strategy of vascular-targeted therapy for liver fibrosis

Tocotrienol in the Management of Nonalcoholic Fatty Liver Disease: A Systematic Review

Summary: The burden of NAFLD is growing, but effective pharmacological treatments are still lacking. Alpha-tocopherol, a vitamin E isomer, is currently recommended for NAFLD patients. Another vitamin E member called tocotrienol (T3) has shown potential in managing NAFLD due to its anti-inflammatory and antioxidant properties. This systematic review found that T3 supplementation improved NAFLD outcomes in both clinical and preclinical studies. However, more comprehensive studies are needed to validate its effectiveness.

Vascular traffic control of neutrophil recruitment to the liver by microbiota-endothelium crosstalk

Summary: Using in vivo imaging, researchers found that the intestinal microbiota guides neutrophils to the liver during bloodstream infections. This process is mediated by the microbial metabolite D-lactate, which enhances neutrophil adhesion in the liver. Gut-to-liver D-lactate signaling prompts liver endothelial cells to upregulate adhesion molecule expression, promoting neutrophil adherence. Targeted correction of microbiota D-lactate production restored neutrophil homing to the liver and reduced bacteremia in a model of Staphylococcus aureus infection.
