Aminopeptidase A inhibitors as centrally acting antihypertensive agents

Among the main bioactive peptides of the brain renin-angiotensin system, angiotensin (Ang) II and AngIII exhibit the same affinity for the type 1 and type 2 Ang receptors. Both peptides, injected intracerebroventricularly, cause similar increase in blood pressure (BP). Because AngII is converted in vivo to AngIII, the identity of the true effector is unknown. This review summarized recent insights into the predominant role of brain AngIII in the central control of BP underlining the fact that brain aminopeptidase A (APA), the enzyme forming central AngIII, could constitute a putative central therapeutic target for the treatment of hypertension. This led to the development of potent, systematically active APA inhibitors, such as RB150, as a prototype of a new class of centrally acting antihypertensive agents for the treatment of certain forms of hypertension.

Summary: Spinal cord injury results in sensory-motor function loss due to damage between the brain and spinal neurons. This leads to reduced locomotor function, respiratory impairments, and increased cardiovascular risk. Pharmacologic neuromodulators, such as 5-HT1A receptor agonists, show potential in inducing functional improvements by activating spared motoneurons. Additionally, combining 5-HT1A agonists with other therapies may be even more promising.

Umbilical cord-derived mesenchymal stromal cell therapy to prevent the development of neurodevelopmental disorders related to low birth weight

Summary: Low birth weight (LBW) increases the risk of neurodevelopmental disorders (NDDs) such as attention-deficit/hyperactive disorder and autism spectrum disorder, and there are currently no prophylactic measures available. Neuroinflammation plays a major role in NDDs, and umbilical cord-derived mesenchymal stromal cells (UC-MSCs) have immunomodulatory properties. This study found that systemic administration of UC-MSCs in the early postnatal period improved hyperexcitability in LBW pups, but the beneficial effects for NDDs were marginal.

Summary: Congenital Central Hypoventilation Syndrome, a rare disease caused by PHOX2B mutation, is associated with absent or blunted CO2/H+ chemosensitivity due to the dysfunction of PHOX2B neurons. Clinical observations have reported non-systematic CO2/H+ chemosensitivity recovery under desogestrel. This study used a preclinical mouse model to investigate the restoration of chemosensitivity by etonogestrel acting on serotonin neurons or residual PHOX2B cells.
