Chemical modified natural nanogels crosslinked with S Benzyl L cysteine exhibit potent antibacterial activity

Improper use of antibiotics has accelerated the global rise of drug-resistant bacteria, especially strains that form biofilms. With the goal of achieving good biocompatibility while reducing adverse effects, we extracted nucleic acid from kiwifruit and used it as a molecular backbone. After HCl treatment, the DNA increased its opened deoxylriboses. S-Benzyl-L-cysteine (SBLC) was then crosslinked with the aldehyde groups in DNA under emulsion processes, becoming SBLC-modified DNA-HCl nanogels. These natural NGs modified with hydrophobic SBLC were analyzed for antibacterial activities. In cell viability assay, these modified natural NGs revealed low cytotoxicity toward normal cells. In antibacterial assays, these modified natural NGs inhibited bacteria, especially biofilms-formed P. aeruginosa and pathogenic E. coli, while exhibiting bacterial membrane rupture as seen through SEM and ameliorating P. aeruginosa-induced lethality in C. elegans. These modified natural NGs may provide a novel anti-biofilm agent in combating resistant bacterial infection.
