Summary: The mechanism of endo-type action on laminarin by GH55 endo-beta-1,3-glucanase MnLam55A was investigated, revealing its preference for cleaving the nonreducing terminal beta 1-3-linkage of the laminar-ioligosaccharide moiety. The crystal structure of MnLam55A showed similarities to exo-beta-1,3-glucosidases, but possessed an extended substrate-binding cleft.

Summary: This study successfully expressed and purified an endo-beta-1,3-glucanase PeBgl1 from Penicillium expansum, and analyzed its characteristics. The PeBgl1 protein showed good enzyme activity at 50-60 degrees C and at pH 5.0-9.0, and it was not metal dependent, making it potentially robust for storage and transportation.

Summary: EFSA was asked by the European Commission to evaluate the application for renewal of Agal-Pro BL/BL-L & REG;. The additive is considered safe for animals, consumers, and the environment under the authorized conditions of use. It is an irritant to the skin and eyes and a dermal sensitiser, and should be considered a respiratory sensitiser due to the proteinaceous nature of the active substances. The efficacy of the additive does not need to be assessed for the renewal of the authorization.

Summary: In this study, NAD(+)-dependent formate dehydrogenase from the thermotolerant yeast Ogataea parapolymorpha DL-1 was overexpressed in E. coli cells and successfully purified. Crystallization experiments revealed that the mutant OpaFDH_AD was able to produce crystals of the holo-form under specific conditions, enabling the collection of X-ray diffraction data.

Summary: DNA sliding clamps, such as PCNA and 9-1-1, play important roles in DNA replication and repair by recruiting proteins involved in these processes. While PCNA and 9-1-1 have similar structures, the mechanism of interaction between 9-1-1 and its partners is distinct from that of PCNA.

Summary: The sliding DNA clamp protein plays a crucial role in cell survival and proliferation and is considered a promising target for bacterial infection therapy. The crystal structure of the bacteria Clostridioides difficile's DnaN protein has been determined, revealing similarities to DnaN from other bacteria but with differences in the binding pocket for partner proteins. These findings provide insight into the development of new therapies for C. difficile infection.
